Applying the ion beam to the Bacteria of Cellulose hydrolysis is a new attempt. At the same time, the research on the Bacteria of Cellulose hydrolysis implanted by low energy ion will further the study on the biological effect of ion beam and widen the application range of ion beam biotechnology in the industrially microbiological screening.Without documents related to original strain, this thesis had to probe into the primary technological conditions of original strain. And eventually made the certain radial fermentation condition.Determination of cellulase activity and reducing sugar by Dinitrosalicylic (DNS) acid reagent were optimized. The physiological character of cellulase and the best culture pH, temperature and stability were quested for. Then according to this base, this thesis analyzed the influence of some parameters, including energy and dose for the purpose of finding the best ion mutant parameters, and studied the mechanism of implanted ion on this strain.The original strain of the Bacteria of Cellulose hydrolysis HY2 was optimized by using ion implantation and then was induced and screening. At last the high-yielded strain HY2106 was acquired. Its Filter Paper Activity (FPA) enzyme and Carboxymethylcellulose (CMCase) enzyme was respectively enhanced 127.8% and 110.2%, and the genetic stability was detected. The decomposable capability of high-yielded strain HY2106 and original strain HY2 was compared.
