Experimental Study On The Intervention Effect And Mechanism Of Yiqi Jianpi Kangai Prescription Combined With Capecitabine On Colorectal Cance

Purpose:With spleen deficiency in colorectal cancer as the focus,I researched ancient and modern literature and summarized clinical experience and principles of treatment.In addition,I explored the theoretical evidence of Yi Qi Jian Pi Kang Ai Formula in colorectal cancer treatment and assessed its clinical efficacy.Lastly,through animal experiment using CT26 WT cancer cell lines transplanted onto naked mice,explore the anti-cancer and protective effects as well as the mechanisms of action of Yi Qi Jian Pi Kang Ai Formula,when complemented with capecitabine,in colorectal cancer treatment,thereby providing evidence for its usage in the treatment of colorectal cancer together with capecitabine.Material and method:This experiment used SPF grade BALB/C thymus knocked-out naked mice as the target of experiment.Firstly,40 mice were used,of which there were 20 male and female mice each.The mice are 6 weeks old with weight of 20±5g.CT26 WT cancer cell lines in 0.2ml suspension was injected beneath the skin of the armpit to create the colorectal cancer mice models.The target for comparison is the model group.Using random number method,the 40 mice were randomized into 4 groups,namely model group,Yi Qi Jian Pi Kang Ai Formula group,western medicine capecitabine group as well as Yi Qi Jian Pi Kang Ai Formula and capecitabine combined group.Subsequently,the 4 groups will receive their respective intervention and after 15 days of intervention,the experiment would conclude and the necessary materials harvested.In additional,the weight of the mice was taken before and after intervention.The general well being of the mice were observed every other day in the course of the experiment.At the end of the experiment,the blood of the mice was harvested for the eventual blood test.Furthermore,the whole cancer body will be harvest and weighed.Lastly,with the use of SPSS 18.0 software,analyzed the data and a P value of <0.05 was set as being statistically significant.Results:A)The impact of Yi Qi Jian Pi Kang Ai Formula on the anti-cancer efficacy as well as the protective efficacy when complemented with capecitabine.1.Weight of the tumor: Yi Qi Jian Pi Kang Ai Formula was able to improve the anti-cancer efficacy of capecitabine and the combined intervention group achieved better anti-cancer efficacy compared to single intervention groups.The single intervention groups.3.NLR:Chinese medicine may enhance the anti-cancer effect of capecitabine by reducing Neu and up-regulating Lym,and resulting in a down-regulating NLR.4.PLR: Chinese medicine may increase the anti-cancer effect of capecitabine by up-regulating Plt and Lym,and resulting in down-regulating of PLR.5.Weight of nude mice with transplanted tumor: The rate at which the weight increase in capecitabine intervention group is lower than that of the other three groups,thus capecitabine has certain side effects on the body.6.Liver and Kidney function test: Capecitabine has exhibited hepatotoxicity to the liver,evident by the increase of AST,ALT and AST/ALT levels,while Yi Qi Jian Pi Kang Ai Formula,when complemented with capecitabine,can reduce the side effects of capecitabine on the liver through its liver protective effects,thus resulting in the decrease of AST,ALT and AST/ALT levels;Capecitabine has exhibited an impact on renal function,which leads to the increase of BUN value.At the same time,the complementation of Chinese medicine can reduce the nephrotoxicity of capecitabine,which leads to the drop in BUN value.7.RBC and Hb result: During the drug intervention,capecitabine did not cause the decrease of RBC and Hb values.B)The mechanism of action of Yi Qi Jian Pi Kang Ai Formula when complemented with capecitabine in the area of anti-cancer1.HO-1,NF-k Bp65 and IL-6 results: The anti-cancer effect of the Chinese medicine prescription for Yi Qi Jian Pi Kang Ai Formula may be achieved by down-regulating the signal pathway of NK-k B/IL-6/JAK/STAT/HO-1,thus enhancing the anti-cancer effect of capecitabine.2.E-cadherin and Vimentin results: The anti-cancer effect of capecitabine is enhanced by down-regulating Vimentin and up-regulating E-cadherin by Yi Qi Jian Pi Kang Ai Formula,and the mechanism of this effect may be related to the mechanism of down-regulating IL-6 in Chinese medicine.Conclusion:1.Yi Qi Jian Pi Kang Ai Formula has displayed anti-cancer effects.In addition,Yi Qi Jian Pi Kang Ai Formula when combined with capecitabine has displayed greater anti-cancer effect.Furthermore,Yi Qi Jian Pi Kang Ai Formula has shown to be able to reduce the side-effects of chemotherapy on the body and thus allowing for better prognosis.2.Yi Qi Jian Pi Kang Ai Formula achieved its effect potentially through the down-regulation of HO-1,NF-k Bp65,IL-6 and Vimentin,while up-regulating of E-cadherin.
