ObjectiveA large amount of research evidence shows that the stemness of cancer cells is the cause of drug resistance or metastasis,and is an important therapeutic target.PRDM14(PR domain zinc finger protein 14,PRDM14)has entered the field of stem cell researchers in recent years.It can regulate the pluripotency of primordial germ cells,is expressed in a variety of malignant tumor cells,and confers stem-like characteristics on primordial tumor cells.Tumor occurrence and metastasis suggest that it may be a new tumor target.E-cadherin and Vimentin are classic markers of epithelial-mesenchymal transition.Epithelial-mesenchymal transition is an important pathway for colorectal cancer to metastasize,and studies have shown that epithelial-mesenchymal transition is also involved in cancer stem cells(CSC)form.This study mainly explored the expression differences of PRDM14 and its relationship with clinicopathological characteristics of patients with colorectal cancer(colorectal carcinoma,CRC)adjacent normal tissues,colon or rectal tubular adenomas,and colorectal cancer tissues;analysis of the colon or the relationship between PRDM14 and E-cadherin,Vimentin in rectal cancer tissue.MethodsCollected 106 cases of colorectal cancer(CRC)patients with cancer tissues and corresponding adjacent tissues(≥5cm)44 cases(62 cases corresponding to adjacent tumors were excluded due to technical reasons),and a total of 45 cases of colon or rectal tubular adenomas were collected.Tissue microarrays were used to detect the expression and distribution of PRDM14 protein in the above 151 cases of benign and malignant intestinal tumors and corresponding 44 cases of adjacent tissues by immunohistochemistry(Streptavidin-peroxidase detection system);using SPSS Statistics 25.0 Analyze and process the data,and finally achieve the purpose of exploring the impact of PRDM14,E-cadherin,and Vimentin on patients with colorectal cancer and the relationship between the threeResults1.The expression rate of PRDM14 in colorectal cancer tissue is 100%(106/106),the expression rate in tubular adenoma tissue is 86.67%(39/45),and the expression rate in adjacent tissues is47.73%(21/44).There are statistical differences.Academic significance(c2=20.133,P<0.001).2.The expression site of PRDM14 protein in the paracarcinoma-tubular adenoma-carcinoma gradually transitions from the nucleus to the cytoplasm,and the difference in the expression site is statistically significant(c~2=9.979,P=0.007).3.PRDM14 protein expression and patient’s gender(c2=5.434,P=0.020),degree of differentiation(c2=76.415,P<0.001),depth of tumor invasion(c~2=19.962,P<0.001),regional lymph node metastasis(c~2=51.245,P<0.001),distant metastasis(c~2=76.415,P<0.001),p TNM staging(c~2=30.075,P<0.001)were significantly related,but not significantly related to the patient’s age and tumor site(P>0.05).4.The expression of E-cadherin protein in colorectal cancer is related to lymph node metastasis(c~2=16.944,P<0.001)and p TNM staging(c~2=17.624,P<0.001),and is related to the patient’s age,gender,tumor location,and degree of tumor differentiation There was no significant correlation between the depth of tumor invasion,lymph node metastasis,distant metastasis,p TNM staging,etc.(P>0.05).5.The expression of Vimentin protein in colorectal cancer is related to lymph node metastasis(c~2=31.648,P<0.001),distant metastasis(c~2=4.987,P=0.026)and p TNM staging(c~2=33.756,P<0.001),which is related to the patient There was no significant correlation between age,gender,tumor location,tumor differentiation degree,and tumor invasion depth(P>0.05).6.The combined detection of E-cadherin and Vimentin found that its expression is related to lymph node metastasis(c~2=39.509,P<0.0001)and clinical stage in patients with colorectal cancer(c~2=41.836,P<0.0001)7.In colorectal cancer,PRDM14 protein expression is negatively correlated with E-cadherin expression(r=-0.219,P=0.024),and positively correlated with Vimentin expression(r=0.519,P<0.001);E-cadherin protein and Vimentin protein expression were negatively correlated(r=-0.358,P<0.001).8.When E-cadherin expression is positive,PRDM14 expression is only related to tumor invasion,and the difference is statistically significant(P=0.002);when E-cadherin is negative,PRDM14 expression is related to tumor invasion,lymph node metastasis,distant metastasis,and p TNM staging,The difference was statistically significant(P<0.05).9.When Vimentin is negative,the expression of PRDM14 is related to tumor invasion and p TNM staging,and the difference is statistically significant(P<0.05).10.Kaplan-Meier survival curve statistical analysis shows that the high expression of PRDM14protein is related to the 5-year progression-free survival(PFS)of colorectal cancer patients(P<0.05).Compared with the high expression of PRDM14,the low expression 5 Annual PFS is higher;Vimentin protein expression is also related to patients with 5-year PFS.Compared with negative patients,positive expression has a worse prognosis;while E-cadherin expression has nothing to do with colorectal cancer patients with 5-year PFS;The degree of differentiation of rectal cancer,distant tumor metastasis,and p TNM staging are all risk factors for poor prognosis in patients with colorectal cancer.Conclusions1.PRDM14 protein expression is increases prior to tumorigenesis in colorectal cancer,which is a risk factor.Through immunohistochemistry analyses of clinical tissues,we detected PRDM14-positive cells in the process of normal tissue adjacent to cancer-adenoma-cancer,the high expression rate of PRDM14 is getting higher and higher,and the expression site gradually transitions from the nucleus to the cytoplasm.2.The expression of PRDM14 protein is related to the gender of colorectal cancer patients.The high expression rate of PRDM14 in male colorectal cancer patients is higher than that in female patients;those with a high degree of differentiation have low expression of PRDM14;those with deep infiltration of the serosal membrane have high expression of PRDM14;The more metastatic areas and the higher the clinical stage,the higher the expression rate of PRDM14;in addition,the prognosis of patients with high expression of PRDM14 is worse than that of patients with low expression.3.The expression of E-cadherin protein is related to regional lymph node metastasis and p TNM staging.However,the positive expression of E-cadherin in this experiment does not affect the prognosis of patients with colorectal cancer.4.Vimentin protein expression is related to regional lymph node metastasis,distant met astasis,p TNM staging,and the prognosis of Vimentin positive expression is worse tha n negative.5.Vimentin protein expression is negatively correlated with E-cadherin protein expression;PRDM14 protein expression and E-cadherin expression are negatively correlated,and Vimentin expression is positively correlated.The state is irrelevant.The combined detection of PRDM14,E-cadherin and Vimentin helps to assess the infiltration of colorectal cancer. |
