| Objective: The clinical observation of the "Guben Peiyuan" acupuncture method on patients with early myopia was evaluated,and the results of the experimental study on the changes of inflammation-related differential proteins and related pathways in the scleral samples were preliminarily explained,and the biological effect mechanism of the "Guben Peiyuan" acupuncture method in the treatment of early myopia was preliminarily explained.Methods:Clinical trial studies1.Baseline period assessment of healthy subjects;2.Effect of "Guben Peiyuan" acupuncture method on relevant indicators of patients with early myopia.Effect mechanism studyThe obtained sclera samples were screened by using "Label-free" proteomics technology,and the differential proteins were identified,and their related molecular functions and related protein pathways were analyzed using KEGG,Inter Pro and GO databases.The differences of myopia-related proteins and the enriched pathways in each group were compared,and the molecular mechanism of the effect of "Guben Peiyuan" acupuncture in the treatment of myopia was discussed.Results:Clinical trial studies1.Baseline assessment of subjects with early myopia: no difference in age and course of the two groups.2.The "Guben Peiyuan" acupuncture method has a certain improvement effect on patients with early myopia.Effect mechanism studyA total of 2309 quantifiable pieces of information were identified in this research.Compared with normal sclera,55 proteins changed significantly in myopic sclera.Compared with the "Guben Peiyuan" acupuncture group before and after treatment,31 proteins changed significantly,and in the conventional acupuncture group,38 proteins changed significantly.significant changes in the protein.Comparing the "Guben Peiyuan" acupuncture group with the control group,there were 15 differential proteins,including 8key proteins.6 main differential protein pathways were enriched in the "Guben Peiyuan" acupuncture group before and after treatment,2 related protein pathways in the control group,and 4 related protein pathways in the conventional acupuncture group.Comparison of the above differences,all p<0.05.Conclusion:1 In this study,the guinea pig with shape deprivation myopia model was successfully constructed;2 In the clinic,the "Guben Peiyuan" acupuncture method and conventional acupuncture improve the naked eye vision of myopic patients to a certain extent;3 In the experiment,the "Guben Peiyuan" acupuncture method and conventional acupuncture had good intervention effects on various indicators of myopia model guinea pigs.4 In experimental studies,we found changes in inflammatory pathways and inflammation-related proteins,suggesting that the mechanism of acupuncture treatment for myopia may be related to inflammatory pathways,laying the foundation for the next experimental verification. |
