ObjectiveSystematically summarize the clinical experience,medication characteristics,core prescriptions and academic ideas of a hospital in the treatment of lung cancer.Based on the core prescription of traditional Chinese medicine in the treatment of lung cancer and using the research method of network pharmacology,this study analyzes the potential mechanism of the core prescription of traditional Chinese medicine in the treatment of lung cancer,provides data reference and basis for the clinical syndrome differentiation and treatment of lung cancer,and provides reference for the research and development of new drugs and their mechanism.MethodsThis study mainly collects and analyzes the electronic medical record information of lung cancer inpatients in a hospital from January2017 to December 2020.With the help of IBM SPS S Modeler 18.0 and Qihuang data AI workstation software system cluster analysis and complex network analysis,this study excavates and summarizes the medication characteristics,core prescriptions and academic ideas of lung cancer treatment in a hospital.At the same time,through data analysis,the core prescription of traditional Chinese medicine in the treatment of lung cancer is analyzed and studied by network pharmacology.Firstly,we make full use of the existing gene bank resources at home and abroad to comprehensively collect the genes related to the pathogenesis and related diagnosis and treatment of lung cancer as much as possible,focus on searching the research information of lung cancer related genes in GWAS Catalog database,OMIM database,Gene Cards database,GEO database and Pubmed literature,and take the gene information with high frequency of lung cancer research as the lung cancer related genes.Secondly,through the pharmacology database of traditional Chinese medicine system,we screen the active ingredients(including the drug components which play a major and auxiliary role in the treatment of lung cancer)of 16 traditional Chinese medicines including : Astragalus,Coix seed,Poria,Coke hawthorn,Virgin,Wolfberry,Bran fried Atractylodes,Mushroom stem,Bran fried citrus husk,Bai Ying,Centipede,Rhodiola,Tiankuizi,Seaweed,Prunella and Shanci Mushroom.All compounds that meet the conditions of oral bioavailability(OB)? 30% and drug class(DL)? 0.18 are used as effective or active components of drugs for the treatment of lung cancer.The Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP)is used to predict and analyze the relevant genes of the screened effective or active components for the treatment of lung cancer and the relevant genes(related to the pathogenesis and diagnosis and treatment of lung cancer)are obtained through the Unitprot database for standardized treatment.Secondly,after merging the genes related to lung cancer(including pathogenesis,diagnosis and treatment)and the genes related to the core prescription for the treatment of lung cancer,we take the intersection of gene data,export the common genes in the core prescription for the treatment of lung cancer,and import the common genes into Cytoscape3.7.2 analyze in the software to construct the phased,correlation and visual network of "drug component target disease" of the core prescription for the treatment of lung cancer.In addition,the screened common genes are introduced into the string database to construct the network diagram of protein interaction and analyzed.Using Cytoscape3.7.2 the built-in cytohubba plug-in in the software selects the core prescription and key genes for the treatment of lung cancer.Finally,the common genes for the treatment of lung cancer in the core prescription obtained through multiple rounds of screening and analysis are imported into David database for gene go and KEGG pathway enrichment analysis,and the analysis results(especially the network association diagram)are visually displayed.ResultsA total of 12191 cases of lung cancer in a hospital meeting the inclusion criteria were collected,including 3234 patients,including 924 female patients(28.57%)and 2310 male patients(71.42%).The number of male patients was 2.5 times that of women.The oldest patient was 88 years old,while the youngest patient was 10 years old and 11 months old,with an average age of 59.02 ± 9.92 years.The age group with the most onset was 55-65 years old,accounting for 35.19%.The specific information is as follows: the treatment of lung cancer in a hospital is mainly warm-property,followed by neutral-property and cold-property medicines.The five flavours are mainly sweet,followed by bitter and Pungent.The meridians mainly involve the lung meridian,the spleen meridian and the liver meridian.Based on Apriori algorithm,the number of co-occurrence of two drugs is 10,and the number of co-occurrence of three drugs is 14.The drugs mainly involved are Astragalus,Poria,Bran Fried Atractylodes,Rhodiola,Coix Seed,Bran Fried Aurantium Shell,Jiao Hawthorn.Through cluster analysis,it is found that traditional Chinese medicine prescriptions are mainly divided into four categories,and drugs can be divided into four categories.The first category(yellow)is Astragalus,Poria,Rhodiola;The second type(red)is Northern Bupleurum,Red peony root,Bran fried Atractylodes,Licorice,Angelica;The third category(green)is Baiying,Ligustrum lucidum,Chinese wolfberry,etc;The fourth category contains more drugs,which can be further divided into 60,classified as A.Dried tangerine peel,B.Curcuma zedoary,Stone primrose grass,etc.,C.Adenophora tetraphylla,Selaginella doederleinii hieron,etc.D.Dried the seed of cowherb,etc.,and others.In addition,using complex network analysis,the core prescription composition of Hubei cancer hospital for the treatment of lung cancer is condensed:Astragalus,Coix seed,Poria,Coke hawthorn,Virgin,Wolfberry,Bran fried Atractylodes,Mushroom stem,Bran fried citrus husk,Bai Ying,Centipede,Rhodiola,Tiankuizi,Seaweed,Prunella and Shanci Mushroom.In the network pharmacological analysis of core prescriptions,37,1131,296,107 and 204 lung cancer related genes were screened from gwascatalog database,genecards database,OMIM database,geo database and Pub Med literature respectively.1382 disease genes were obtained after integration and elimination of duplication.83 total active ingredients were screened from the core prescription,corresponding to 262 genes.Ten core targets of core prescription for the treatment of lung cancer were screened through protein interaction network,which were VEGFA,AKT1,Jun,PTGS2,CASP3,TNF,IL6,IL1 B,MMP9 and EGF.Through go enrichment analysis,it is found that the biological processes involved mainly include: cytokine-mediated signaling pathway,response to drug,positive regulation of gene expression,response to xenobiotic stimulus,Aging,response to hypoxia,positive regulation of transcription,DNA-templated,etc.Through KEGG enrichment analysis,it was found that the active components of various drugs in the core prescription have a certain regulatory effect on Pathways in cancer,AGE-RAGE signaling pathway and Chemical carcinogenesis-receptor activation.ConclusionsAccording to the data mining results,the clinical experience and core prescriptions of a hospital in the treatment of lung cancer are sorted out,and the academic ideas of a hospital in the treatment of lung cancer are summarized:(1)the pathogenesis of "deficiency of vitality" runs through the beginning and the end;(2)It is supplemented with traditional Chinese medicine with anti-tumor specificity to dispel evil and strengthen health.In addition,the data mining research on the drug use law of lung cancer under the background of big data at the macro level is linked with the research on the molecular action mechanism of the core prescription of lung cancer treatment at the micro level,and the clinical data of traditional Chinese medicine in the treatment of lung cancer are analyzed in multiple dimensions,in order to provide some reference for the summary of clinical experience of traditional Chinese medicine in the treatment of lung cancer,the discovery of new drugs and clinical medication.
