Objective On the basis of taking advantage of the good application of deer bone in the past,combined with the unique composition and pharmacological effects of deer bone,a bone repair material based on natural deer bone was developed,and the deer bone was functionalized to have certain antibacterial and antitumor capabilities.To evaluate its application potential in the treatment of orthopedic diseases.Methods 1 ? In this study,the physical dry ball milling method,high temperature calcination method and modified Urist method were used to prepare nano deer bone powder(nBM),nano calcined deer bone powder(nCBM)and nano demineralized bone matrix(nDBM).Biosafety.The biosafety of the prepared nano-bone powder was tested.To evaluate the effects of different bone meal on the proliferation,adhesion,spreading,migration and differentiation of bone marrow mesenchymal stem cells(BMSCs).2?In this study,high-voltage electrostatic drop spheres and phase inversion methods were used to prepare nBM/PLGA microspheres loaded with different proportions(5%,10%,20%,40%)of nano-deer bone powder(nBM).The best bone repair materials were screened out through in vitro cell experiments and in vivo animal experiments,and their potential application prospects in bone repair were evaluated.3?In this study,nBM was used as the substrate material to carry Cu ions to prepare nBM-Cu complexes.To evaluate the antibacterial and antitumor properties of nBM-Cu complexes,develop a new antibacterial and antitumor nanomaterial,and evaluate the application potential of this material in the treatment of bone tumors.Results 1?In this study,nBM,nCBM and nDBM were successfully prepared by different methods,and through the analysis of the basic components of different bone meal,it was found that the calcium-to-phosphorus ratio in deer bone was significantly higher than that of bovine bone and human bone tissue,and it contained bovine bone.Beneficial trace elements such as potassium,iron,selenium,zinc.The three kinds of deer bone powder have good biocompatibility and meet the implantation standard of medical biomaterials.nBM has excellent performance in the proliferation,adhesion,migration and differentiation of BMSCs,and was used for follow-up research.2?In this study,osteogenic microspheres with different proportions(5%,10%,20%,40%)of nBM/PLGA were prepared by high-voltage electrostatic drop spheres and phase inversion method.In vitro cell biological function studies showed that 20%nBM/PLGA microspheres and 40%nBM/PLGA microspheres had better effects on promoting the proliferation and osteogenic differentiation of BMSCs.The results of in vivo skull defect repair experiments showed that 20% nBM/PLGA microspheres had faster mineralization and the ability to promote new bone formation.3?In this study,a polymer fiber-metal complex(nBM-Cu)was successfully constructed using nBM as the substrate material as a catalyst for the heterogeneous Fenton reaction.Compared with the neutral environment,the nBM-Cu complex has higher catalytic efficiency under acidic conditions and generates more active hydroxyl radicals.Since the concentration of H2O2 in tumor cells is higher than that of normal cells,and the tumor microenvironment is more acidic than normal tissues,nBM-Cu can selectively kill tumor cells.In addition,due to the coexistence of the antibacterial ability of copper ions and the dual sterilization mechanism of reactive oxygen species related to nBM-Cu,nBM-Cu exhibits a good bacterial killing effect and has a certain potential as an antibacterial material.Conclusion 1 ? The nBM,nCBM and nDBM prepared in this study have good biocompatibility,and nBM has excellent performance on the proliferation,adhesion,migration and differentiation of BMSCs.2?The high nBM content nBM/PLGA microspheres prepared in this study have potential applications in bone tissue engineering and stem cell expansion.3?The nBM-Cu complex prepared in this study provides a new strategy for bone tumor treatment and infection prevention.Due to its good tumor inhibitory effect and antibacterial properties,it has certain potential in the clinical application of bone tumor therapy.
