Objective:1.Explore the correlation between the hot and humid asthma rat model under the guidance of traditional Chinese medicine theory and the modeling of different phenotypes of asthma in modern medicine;2.Explore the therapeutic mechanism of Jiawei Yinchenhao decoction in rats with damp-heat asthma;3.Explore the correlation between the acute attack period of children's damp-heat asthma and common clinical physical and chemical indicators.Methods:Experimental study:1.The rat model of damp-heat asthma was established by using high-fat feed+long-term exposure to damp-heat environment+ovalbumin sensitization and multi-factor combined intervention.The rat model with simple syndrome of damp and heat was established by using high-fat feed+long-term exposure to humid and hot environment and combining the internal and external wet factors.The asthmatic rats were compared with normal rats and ovalbumin sensitized rats.2.Experi-mental rats were randomly divided into 6 groups:blank group,model control group,dexamethasone group,and low-dose,medium-dose and high-dose groups of Jiawei Yinchenhao decoction.The general state of rats in each group was recorded,and the cell count of alveolar lavage fluid and the expression of IL-8,IL-17,serum HDL,LDL,plasma FIB,lung histopathology,and ROR-?t m RNA in lung tissue were detected,so as to explore the therapeutic effect and mechanism of Jiawei Yinchenhao decoction on dampness-heat asthma.Clinical research:The clinical case review analysis table was developed,and the relevant information was statistically described for the cases that met the standards.Besides,the microscopic syndrome differentiation indexes of children with damp and heat asthma were explored through Logistic regression analysis and ROC curve analysis.Results:1.The proportion of NEU in alveolar lavage fluid in the humid heat asthma model was significantly higher than that in the traditional"pure OVA asthma model";The simple model of"internal and external humidity"showed obvious airway inflammatory response.2.Jiawei Yinchenhao decoction can improve the symptoms of dampness-heat syndrome and asthma in rats with damp and heat asthma model,up-regulate HDL level,down-regulate LDL and HIB levels,and improve airway inflammation.It can significantly reduce the expression of ROR-?t m RNA in lung tissue,and down-regulate the levels of IL-8 and IL-17,and down-regulate the proportion of neutrophils in alveolar lavage fluid.3.The incidence of damp heat asthma was mostly in boys,with high incidence in pre-school age children.Most hospitalized children were in the acute attack period of asthma.Hot and humid asthma was more common in autumn and less common in winter,which showed a statistically significant difference with non-hot and humid asthma(P<0.05).Po-sitive epstein-barr virus test was not significant for the difference between damp and heat asthma(P>0.05).The positive MP test in children with asthma indicated that they were of damp and heat asthma,which was statistically significant compared with non-damp and heat asthma(P<0.01).The eight respiratory etiology tests indicated that the positive rate of damp and heat asthma virus was higher.4.WBC,L,M,N,EO,BASO,rdw-cv,PLT,PDW,CRP,NLR,PLR,ELR,ENR,EMR,NMR,MLR,K,Na,Cl,Ca,CO2,ALT,AST,ALP,UREA,Ig E,?2MG,CK-MB,LDH,ESR,GLU had no statistical significance in the auxiliary diagnosis of damp heat asthma(P>0.05).The elevation of RBC,HGB,BLR,TP,TBIL and CREA was statistically significant for the auxiliary diagnosis of damp-heat asthma(P<0.05).Conclusion:1.The damp-heat asthma rat model established by high-fat and high-protein feed+intermittent damp-heat environmental exposure+ovalbumin sensitization can be used as a new neutrophilic asthma model;2.The combined intervention of"internal and external dampness"with high-fat and high-protein feed and intermittent exposure to hot and humid environment can lead to significant airway inflammatory infiltration in rats.3.Jiawei Yinchenhao decoction can reduce the expression of ROR-?t m RNA in lung tissue,down-regulate the levels of IL-8 and IL-17,and reduce the accumulation of airway neutrophils.The therapeutic mechanism may be related to the Th17 cell pathway.4.There was a certain correlation between damp-heat asthma and autumn onset,MP infection and increased detection of RBC,HGB,BLR,TP,TBIL and CREA.
