Functional dyspepsia (FD) is a common disorder of gut motility in clinic. FD is a most important topic in gastrointestinal disease research globally. To date, the prokinetic effect of western medicine is far from satisfactory due to side effects. Some traditional Chinese medicines (TCM) have produced relatively favorable effect to FD in clinic. This research focuses on the Chinese materia medica Fructus aurantii (FA), Fructus aurantii and Magnolia bark decoction (FM), Xiaoyaosanjiawei decoction (XSJ) and Merazin hydrate (MH) which was recently issolated from FA. We study on the regularity of FA on gastrointestinal motility and the prokinetic effect and mechanism of MH.Objectives:study on the regularity of FA on gastrointestinal motility and the prokinetic effect and mechanism of MH.Methods:(1) we provide a simple, reliable and rapid method for simultaneously quantitating the five compounds (narirutin, hesuy peridin, naringin, neohesperidin and merazin hydrate) in FA-type decoction. (2) we provide a simple, reliable and rapid method for simultaneously qualitating the five compounds (narirutin, hesperidin, naringin, neohesperidin and merazin hydrate) in jejunum after oral administration of FA decoction to rats for the first time. (3) Investigate the prokinetic effect of FA, FM and XSJ in vivo and in vitro. (4) Because the effects of four flavonoids in FA have been reported for many times and the function of MH has been little studied, we will investigate the effect of MH on intestinal motility and its prokinetic mechanism.Results:The amounts of merazin hydrate, narirutin and neohesperidin were in the order FM>XSJ>FA, hesperidin and naringin were XSJ>FM>FA. The five compouds are the main components in FA. They also can be absorbed into jejunum after oral administration of FA decoction to rats. FA and FM can promoted intestinal transit and gastric emptying in high doses and inhibit the gastrointestinal motility in low doses. XSJ also have prokinetic effect in low dose. MH can promote intestinal transit and gastric emptying in a dose-dependent manner when gavaged acutely and it was, at least in part, mediated by stimulation of H1 histamine receptors. It can also significantly reverse cisplatin-induced delay in gastric emptying.Conclusions:FA and FM FA and FM can promoted intestinal transit and gastric emptying in high doses and inhibit the gastrointestinal motility in low doses. XSJ also have prokinetic effect in low dose. MH can promote intestinal transit and gastric emptying in a dose-dependent manner and it was, at least in part, mediated by stimulation of H1 histamine receptors. It can also significantly reverse cisplatin-induced delay in gastric emptying. The research of the prokinetic effect of MH can provide an experimental basis for new drug development.
