Research On Complex Network Based On Data Mining For Phlegm-dampness Constitution Differentiation And Treatment

Objective:In this study,I focus on the constitutional differentiation and treatment to phlegm-dampness constitution,by overviewing the the modern literature of constitutional differentiation and treatment of phlegm-dampness constitution and related-diseases.To research constitutional differentiation and treatment of phlegm-dampness constitution by arranged and digged the current clinical data with the literature research method,data mining method,prediction method of complex network,evidence-based medicine method.To predict the possible mechanism of phlegm-dampness constitution by summed up the current clinical treatment characteristics and drugs on this constitution and from the perspective of drug network targets and pathways.To evaluate the efficacy of traditional Chinese medicine in the health management of phlegm-related chronic diseases,and to summarize the safety and effectiveness of traditional Chinese medicine in health management.Method:The thesis consists of three parts:1 In literature research,analysis of the theory of style herbs treatment of phlegm-dampness constitution in the literature,through data mining,summed up the characteristics of the herbs,using visualization of complex network analysis software to framework herbs target-disease network construction.2 Pathway,target,and disease network were constructed by network pharmacology.Based on the bioinformatics analysis,the data mining results were analyzed by Pathway,GO,OMIM and TTD,to predict the pathogenesis of phlegm-dampness constitution..3 System review and meta-analysis of the effectiveness and safety of traditional Chinese medicine intervention in phlegm-dampness chronic diseases in health management.Result:1 There were 42 literature in first part,phlegm-dampness constitution differentiation and treatment by traditional Chinese medicine combined with lifestyle intervention in the 28 articles,prescription of traditional Chinese medicine decoction to mainly use 99 kinds of commonly used traditional Chinese medicine drugs(frequency>10),association rules analysis of drugs,founded 53 drug association rules of correlation analysis results show that a variety of drugs,drug combination of Poria,Atractylodes,pinellia,dried orange peel is the core of the drug.Construction of drug target disease network using R software.2 Poria,Atractylodes,pinellia,dried orange peel to construct a network KEGG pathway enrichment analysis and GO analysis,OMIM,TTD disease enrichment analysis,analysis results showed that:these four drugs tagets included opioid(OPR)and glucocorticoid receptor(N3RC1)in neuroactive ligand-receptor interaction pathway,acetylcholine receptor in Ca2+pathway of the cell membrane.The four drugs were mainly involved in the pathway of fatty acid degradation,gluconeogenesis and glycolysis.The four drugs have a close relationship with neuroendocrine,which is enriched in the central nervous system neurotransmitter acetylcholine,GABA,5-HT.3 GO analysis showed that the four drugs were enriched in the body fluid circulation,cell-cell signaling,aging,lipid metabolism,redox activity,cell proliferation,cell apoptosis,cell migration,cell membrane protein changes and so on.It is mainly involved in the metabolism of amino acid,protein,metabolism of small molecules,the encapsulation of membrane and the transport of transmembrane proteins.4 Analysis the results through OMIM and TTD database,showed that the four drugs were associated with Alzheimer's disease,cancer,mental disorders,endocrine and metabolic diseases,pain,asthma,stress disorder,benign prostatic hyperplasia,heart failure and other diseases.5 Complex network analysis showed that the four drugs have 13 targets in common,these targets included ANXA1,AR,PGR,CD34,FGFR2,MED1,NR3C1,OPRK1,PGR,TNF,TRIM24,TRPA1,TRPV3.Which has anti-inflammatory,anti-tumor or,cell cycle related targets are ANXA1,TNF,and TRIM24,endocrine and metabolic target is AR,PGR,PGR,NR3C1,OPRK1,and feel the chemical and physical stimulation related TRPA1,TRPV3.6 In 38 studies involved in the assessment of efficacy of TCM different whether the use of Chinese medicine in the treatment of chronic diseases in 3241 patients,the quality of the literature included in the study,meta analysis showed that every two types of comparison of various outcome measures showed that the effect of the use of traditional Chinese medicine is better than that without the use of traditional Chinese medicine,whether it is only by health management or health management combined with conventional western medicine therapy,there are more effective with Chinese medicine.In these 38 studies,there were 21 studies used the four herbs,Poria cocos,Atractylodes macrocephala,Pinellia ternate and tangerine peel,of which 13 used four herbs,3 herbs were used in 8 studies.Conclusion:In the first part,in the constitutional differentiation and treatment of phlegm dampness constitution related chronic diseases,by the way of guide the lifestyle combined with herbs with TCM constitution theory as the main method,the diseases were mainly obesity,insulin resistance,type 2 diabetes and hypertension as the core of the metabolic syndrome,the clinical research results show that the Constitution of TCM syndrome based differentiation can to achieve effective prevention and treatment of these diseases.the law of Phlegm dampness syndrome should insist on the invigorating the spleen for eliminating dampness and phlegm turbidity,most of the current clinical prescription were self-prescription;from the efficacy of drugs,the most used is the tonic medicine,from the drug efficacy point of view,the most used is tonic medicine,followed by Diuresis moisturizing drugs,phlegm cough and asthma drugs,blood circulation drugs.The drug properties are mainly warm,and the use of cold and hot drugs were less.The five flavors were sweet,bitter and pungent.Drug owned by the spleen,lung,stomach,liver.Through the analysis of the association rules,it was determined that Poria,Atractylodes,Pinellia and Cucumber were the core of the drug for the treatment of phlegm dampness.The complex network analysis was used to form the combination rule of the drug,the angle medicine and the Fang Yuan.The relationship between these drugs suggests that these four drugs may also be the main node of the drug;angle drug network relationship can be seen to the four herbs as the core of the relationship between the drugs;the combination of the four drugs formed the Fang Yuan for the treatment of phlegm constitution.The phlegm dampness constitution differentation under the guidance of the constitution of TCM clinical treatment for lifestyle development may form a new angle of medicine,medicine,but also the formation of a new party element,further research can be carried out from the perspective of prescription compatibility.In the second part,through the analysis of the complex network,four kinds of common drug targets enriched in the body fluid circulation,lipid metabolism,cell signaling,senescence,redox activity,cell proliferation,apoptosis,cell membrane protein,mobile change process.It is mainly involved in the metabolism of amino acid,protein,metabolism of small molecules,the encapsulation of membrane and the transport of transmembrane proteins.Four drugs are associated with Alzheimer's disease,cancer,mental disorders,endocrine and metabolic diseases,pain,asthma,stress disorders,prostate hyperplasia,heart failure and other diseases.The target of the four drugs is acetylcholine receptor and glucocorticoid receptor,which affect the calcium channel.According to the analysis,the target of the treatment of phlegm dampness constitution is the glucocorticoid receptor and TRP family protein.In the third part,the results of the study showed that under various control conditions are the same,for health management of chronic disease,the use of traditional Chinese medicine in various indicators of effect are better than not,therefore,can prompt the clinical use of BianTi disease-diagnosis and treatment method for treatment of patients,at the same time,better effect of combined use of advice the traditional Chinese medicine on the basis of conventional treatment.
