The Study On The Experience Of Professor Xu Li Treating Advanced Lung Cancer Based On Data Mining And The Antitumor Role Of Couplet Medicines

Objectives:To research the medication and compatibility of traditional Chinese medicine prescriptions in the treatment of advanced lung cancer by Professor Xu Li base on data mining.Summarize the experiences in lung cancer treating of Pro.Xu who had lots of experiences in cancer treating especially with traditional Chinese medicine.We also devoted to study the the antitumor role of traditional Chinese medicine Couplet Medicines usually used in the treatment of advanced lung cancer by Professor Xu Li with vitro experiments,which can provide certain theoretical basis for the the mechanism of action of the Couplet Medicines and exploring new direction of advanced lung cancer treatment.Method:We analysed a cohort of 88 advanced lung cancer patients who were treated by Professor Xu Li from January 2016 to October 2018.All of them had been treated by Pro.Xu Li at least three times.The patients general information,Chinese medicine diagnosis and treatment information,Blood examination,imaging examinations,and so on were recorded by the students of Pro.Xu Li in the computer.After removing some useless interference information,we use Medcase V3.2 software to carry out frequency analysis,clustering analysis and association rule analysis on the information of syndromes,tongue and pulse,pathogenesis and prescriptions in medical records.Different concentration of Mao Zhua Cao(Ranunculus ternatus Thunb.)and Jiang Huang(Curcuma longa L.)decoction were add to the Lewis lung cancer cells,We had observed the changes of cell proliferation,cell cycle and cell apoptosis in lung cancer cells and tested the expression of Bcl-2,Caspase 3 and GAPDH mRNA.Result:(1)88 patients with advanced lung cancer,55 males,age ranges from 42 to 79.33 females,age ranges from 42 to 79.33 were squamous cell carcinomas and 55 were adenocarcinomas.(2)Syndrome classification in 88 patients,there were 18 cases of deficiency of Qi and yin,accounting for 20.45%;15 cases of phlegm-heat obstruction of lung,accounting for 17.05%;13 cases of deficiency of qi,phlegm-dampness,accounting for 14.77%;11 cases of deficiency of yin and internal heat,accounting for 12.50%;9 cases of deficiency of lung and spleen,accounting for 10.23%;9 cases of excessive heat toxin,accounting for 10.23%;7 cases of deficiency of lung-yin,accounting for 17.95%;and 6 cases of stagnation of blood and stasis syndrome,accounting for 6.82%.(3)The main clinical symptoms were cough(207 cases,78.23%),shortness of breath(146 cases,55.3%),pleural effusion(131 cases,49.68%),white sputum(87 cases,33.07%)and chest pain(44 cases,16.6%).The main systemic symptoms were fatigue(234 cases,88.5%),poor sleep(193 cases,73.19%),poor appetite(183 cases,69.38%),dry mouth and pharynx(138 cases,52.13%),palpitation(83 cases,31.59%).(4)The results of frequency analysis showed that the top 30 Chinese medicines with high frequency were:Mao Zhao Cao,Jiang Huang,Sheng Huang Qi,Chao Bai Zhu,Fu Ling,Zhu Ling,Tai Zi Shen,Gou Qi Zi,Shi Jian Chuan,San Qi Fen,Sheng Yi Ren,Xian He Cao,Fa Ban Xia,Jie Geng,Zhe Bei Mu,Wu Wei Zi,Shan Yu Rou,Dang Gui,Chen Pi,She She Cao,Bu Gu Zhi,Zi Yuan,Chao Bai Shao,Nu Zhen Zi,Hong Jing Tian,Hong Dou Shan,Ze Qi,Huang Jing,Zhi Gan Cao,Mai DongMao Zhua Cao.(5)Totally 264 prescribtions were selected which involved 289 herbal medicine,the majority were distributed to liver meridian(61.0%),lung meridian(44.4%),stomach meridian(38.6%),spleen meridian(33.6%)and kidney meridian(30.9%)etc in channel tropism.(6)Totally 264 prescribtions were selected which involved 289 herbal medicine,mostly presented sweet(46.0%),bitter(50.2%)or hot(38.1%%)in taste,cold(35.99%)or warm(32.18%)inproperty.(7)In order to find the Couplet Medicines combination of positive strong correlation association rules,after eliminating the rule of promotion in 290 rules and deleting the rule of duplicate Couplet Mediciness,168 Couplet Medicines combinations with strong correlation association rules were obtained.Among them,the rule with the highest degree of promotion was Mao Zhao Cao-Jiang Huang.The Couplet Medicines combination of the rule had the promotion degree of 8.304,the confidence degree of 94.12%,and the rule supports it.The degree of confidence was 10.67%.There were five pairs of regular medicines with 100%confidence,namely:Jiang Huang-Zhu Ling、Jiang Huang-Fu Ling、Dang Gui-Sheng huangqi、maozhuacao-zhuling、maozhuacao-fuling、zisuzi-zisugeng.its regular support degree were 13.21%,13.11%,10.78%,10.67%,9.89%,17.66%,17.87%,12.54%,and the degree of lift was 2.239%,respectively.2.1 74%,2.174%,8.304%,2.467%,2.084%,2.024%,6.786.(8)The concentration of the drug was positively correlated with cell killing,the cell kill rate increased with the increase of drug concentration.There was no significant difference in the apoptotic rate between the high dose group and the cisplatin group(P>0.05).(9)With the increase in the dose of the Mao Zhao Cao-Jiang Huang,the cell growth inhibition increased with the increase of drug concentration.There was no significant difference in cell growth inhibition ability between the high-dose group and the cisplatin group(P>0.05).(10)With the increase in the dose of the Mao Zhao Cao-Jiang Huang,the cells mainly stay in the G1 phase.(11)With the treatment of Mao Zhao Cao-Jiang Huang,the doses of p-p65 and caspase 3 increased,and the expression decreased.Conclusion:Professor Xu Li had a lot of experience in treatment of advanced lung cancer.Base on data mining,accurately analyzed the academic thoughts and prescription rules of dialectical treatment for patients with advanced lung cancer.It can Provide important clinical dialectical ideas and medication for TCM diagnosis in treatment of lung cancer.The prescription had great significance for the promotion of the academic thinking of famous Chinese medicine practitioners.Professor Xu Li’s most commonly used drug in the treatment of advanced lung cancer was the entry point of Mao Zhao Cao-Jiang Huang.Modern research methods have been used to prove that Mao Zhao Cao-Jiang Huang can effectively inhibit the proliferation and growth of tumor cells and promote tumor cell apoptosis.Professor Li is pharmacologically based on the treatment of advanced lung cancer.We had summarized the experiences in lung cancer treating of Pro.Xu Li base on data mining,and studie the the antitumor role of traditional Chinese medicine Couplet Medicines usually used in the treatment of advanced lung cancer by Professor Xu Li with vitro experiments.It provided certain theoretical basis for the the mechanism of action of the Couplet Medicines and exploring new direction of advanced lung cancer treatment.
