Objective: This study is to verify the hypothesis of “the medicinal flavor can be split and composable” via studing the effect of astragalus and its separated components combined with pepperweed seed compatibility on the rat model of harmf?l fluid retention in the upper jiao.And it is also to explore the mechanism of the effect of astragalus split compositions combined with pepperweed seed on the heart and lung function of the rat model of harmf?l fluid retention in the upper jiao.Method: 1.The rat model was established by subcutaneous injection of isoproterenol and endotracheal tube in the scap?lar area.After the rat model successf?lly induced,the astragalus split compositions,the compatibility of astragalus and pepperweed seed,and the compatibility of astragalus split compositions and pepperweed seed were administered to the rats for two weeks in different groups.We explored the effect of astragalus split compositions combined with pepperweed seed on the heart and lung function of the rat model of harmf?l fluid retention in the upper jiao by analyzing the rat body weight(BW),heart weight index(HWI),left ventric?lar mass index(LVWI),lung weight index(LWI),left ventric?lar ejection fraction(LVEF),left ventric?lar fractional shortening(LVFS),plasma creatine kinase(CK),the lung permeability index,the p?lmonary alveolus pumpback amount and the p?lmonary dry wet ratio(W/D).2.By means of ELISA,Western Blotting and Real-Time PCR detection technology,this study explored the mechanism of the effect of astragalus split compositions combined with pepperweed seed on the heart function of the rat model of harmf?l fluid retention in the upper jiao by comparing the expression of Ang ?,AT1,ERK1 / ERK2,CTGF amongdifferent groups' rats in the heart.3.This study explored the mechanism of the effect of astragalus split compositions combined with pepperweed seed on the lung function of the rat model of harmf?l fluid retention in the upper jiao via ELISA,Western Blot and IHC detection technology,by comparing the expression of AQP5?P-CREB and NF-?B P65 among different groups' rats in the lung,Res?lts: 1.Compared with the usage of astragalus,the effects of astragalus split compositions are more significant.The astragaloside has the most obvious effect.The separability of astragalus flavor was verified.Compared with the usage of astragalus and pepperweed seed alone,the effect of astragalus and pepperweed seed compatibility is more significant.The composability of astragalus and pepperweed seed is verified.Compared with the usage of astragalus and pepperweed seed compatibility,the compatibility of astragalus split compositions and pepperweed seed can obviously increase the weight of model rats,improve LVEF and LVFS,reduce heart index,lung index and CK levels in the blood plasma,increase the p?lmonary alveolus pumpback amount and reduce lung permeability index and the p?lmonary dry wet ratio(W/D)significantly.The compatibility of astragaloside and pepperweed seed has the most obvious effect.The combination of Astragalus flavor components and pepperweed seed was verified.2.The compatibility of astragalus split compositions and pepperweed seed can cut in model rats AngII levels in plasma and the myocardial AngII,AT1 receptor mRNA and protein expression of CTGF and can also make the phosphorylation of ERK1/2 level recovered.The compatibility of astragaloside and pepperweed seed has the most obvious effect.In addition,there was a statistically significant difference between the compatibility of astragaloside and pepperweed seed and the compatibility of astragalus and pepperweed.3.The compatibility of astragalus split compositions and pepperweed seed can cut model rats TNF alpha content in plasma and the NF-kappa B p65 protein expression in the lung tissue and can also improve the phosphorylation level of CREB,increasing the expression of AQP5.The compatibility of astragaloside and pepperweed seed has the most obvious effect.Conclusion: The compatibility of astragalus split compositions and pepperweed seed can improve the heart and lung function of the rat model of harmf?l fluid retention in the upper jiao.The mechanism may,on the one hand,reduce the phosphorylation level of erk1/2 and the expression of CTGF by inhibiting the activation of the RAAS in the rat model,on the other hand,cut the TNF alpha content in plasma of model rats and restrain the NF-kappa B pathway,reducing the NF-kappa B in the nuclei of p65 phosphorylation levels.Thus reduce water retention in rat lungs by improving the expression level of p-CREB(Ser133)and AQP5 in the lung tissue.
