Study On Pharmacological Effects Of Semen Pharbitidis And Its Chemical Split Compositions In Rats With Membranous Nephropathy

Semen Pharbitidis is the dry and mature seed of Pharbitis nil(L.)Choisy or Pharbitis purpurea(L.)Voigt in the Convolvulaceae family.It is cold in nature and bitter in flavor.It can cause watery diarrhea to be laxative,tap stagnant with insecticide and dissolve phlegm and flush rheum.Clinically,it is used for the treatment of fecal and urinary stoppage,edema puffiness,retention of phlegm and cough,etc.Ming Yi Bie Lu and many Herbal medicine works records:"semen pharbitidis main gas,cure the foot edema full".In clinical it is used used to treat chronic nephritis edema,has achieved good results.And membranous nephropathy is one of the most common pathological types of the chronic nephritis,which is often characterized by high edema clinically.According to our prophase established extraction and separation technology,Semen Pharbitidis water-decocted was splited into four compositions,namely polysaccharide composition,resin glycoside composition,phenolic acid composition and fatty oil composition.On this basis,combined with the clinical application of Semen Pharbitidis,we carried out the experimental research of Semen Pharbitidis and its chemical split compositions on treatment of rats with Membranous Nephropathy(MN)in this dissertation.This research builds MN model of rats by Cationic Bovine Serum Albumin(C-BSA).Through the observation of the rats with 24-hour urine protein,blood biochemistry indicators(Scr?BUN?TG?TC?TP?ALB),serum antioxidant indicators(MDA?SOD?GSH-PX),and pathologic changes of renal tissue were observed with by using light microscopeand electron microscope,using immunohistochemical staining to check the expression of IL-6?TGF-?1 in the renal tissue,discussing the effect and probable mechanism of semen pharbitidis and its chemical detached compositions in rats with MN.The results showed that Semen Pharbitidis water-decocted,polysaccharide composition significantly decreased MN rats' 24-hour urine protein excretion,reduce the contents of Scr?BUN?TC and TG,increase the contents of TP and ALB,relieve renal pathological injury,reduced the expression of IL-6?TGF-?1 in the renal tissue of MN rats by using immunohistochemical methods.The present study suggests that Semen Pharbitidis water-decocted can improved MN rats' renal function,improved rats hyperlipidemia,reduce the contents of hypoalbuminemia,relieve renal pathological injury,is effective effect in rats with MN.Effective constituents in Semen Pharbitidis treatment of MN may be Semen Pharbitidis polysaccharide components.Semen Pharbitidis polysaccharide components can eliminating oxygen free radicals and antilipoperoxidation,relieve renal injury;inhibit inflammation reaction by expression of IL-6?TGF-?1 in renalt issue of MN rats,inhibit ECM deposit,hold the progression of disease,to achieve the therapeutic effect of MN.In summary,based on the previous studies,this dissertation has explored the the effect of Semen Pharbitidis and its chemical split compositions on treatment of rats with MN in animal experiments,clear the effective constituents and probable mechanism in semen pharbitidis treatment of MN,but also further deepen the understanding of semen pharbitidis effect and it can provide theoretical guidance and reference to further develop clinical application of Semen Pharbitidis and its effective components.
