Objective:Observe the influnce that Xiao Fu Tong Jie Fang treat functional constipation rats from the view of aldosterone ' s synthesis and secretion and the aquaporin water channels.From the view of the kindney adjust water metabolism,we reveal the mechanism of action of nursing functional constipation from the kindey.Methods:1.60 SDrats were randomly divided into 2 groups:blank control group(10rats),functional constipation model group(50 rats).Model groups were given compound diphenoxylate 15mg/kg/ day gavage for 6 weeks,and then colonic tissue were observed with the use of HE staining to verify the models.2.After the models were established,then treated with Xiao Fu Tong Jie Fang.Positive drug group treated with Cong Rong Tong Bian oral liquid.All for more than one months,and then executed.The protein expression of the Aldosterone(ALD)?Renin(PRA)?Angiotensin?(Ang?)?Angiotensin-converting enzyme(ACE)?Arginine vasopressin(AVP)?Cyclic adenosine monophosphate(c AMP)by enzyme linked immunosorbent assay(ELISA).The m RNA and protein expression of MR/AQP1/AQP2/AQP3 in kindney and AQP1/AQP2/AQP3 in colon were determined by Real-time PCR and immunofluorescence assay respectively.Western-Bolt were used to detect the expression of AQP1/AQP2/AQP3 in kindney and colon.3.Using SPSS20.0 software to analysis on the experimental data.Compared among groups and use the single factor variance analysis,when p < 0.05,the difference was statistically significant.Results:1.Xiao Fu Tong Jie Fang affected rat's general physiological condition and the water content of the colon:Compared with the blank control group,the rats of model group had less feces and texture was hard.The water content of the colon levels were significantly decreased(P<0.01).Compared with the model group,the rats of treatment groups had more feces and texture was loose.The water content of the colon levels were significantly increased(P<0.01).2.Xiao Fu Tong Jie Fang affected rat'saldosterone's synthesis and secretion:(1)The influence of Renin(PRA)?Angiotensin?(Ang?)?Angiotensin-converting enzyme(ACE)in serum:Compared with the blank control group,model group,PRA?Ang??ACE content were increased,there was a statistically significant(p<0.05);After administration of the intervention,Xiao Fu Tong Jie Fang each dose group and positive drug group can lower their content,and to reduce the most obvious,a significant difference(p<0.05).(2)The influence of aldosterone's synthesis and secretion:(1)Compared with the blank control group,model group,ALD content were increased(p<0.05);After administration of the intervention,Xiao Fu Tong Jie Fang each dose group and positive drug group can lower their content,and to reduce the most obvious,a significant difference(p<0.05);(2)Compared with the blank control group,model group,MR were the most strongly expressed both in cell membrane and kytoplasm of renal collecting duct.After administration of the intervention,Xiao Fu Tong Jie Fang each dose group and positive drug group,MR were more less strongly expressed both in cell membrane and kytoplasm of renal collecting duct;? MRm RNA relative expression levels were significantly increased(P<0.05);After giving Xiao Fu Tong Jie Fang,MRm RNA expression levels reduced compared with the model group were significantly different(P<0.05).3.Xiao Fu Tong Jie Fang affected rat's Aquaporin water channels:(1)The influence of AVP and c AMP:Compared with the blank group,model group,Levels of AVP?c AMP relative expression levels were increased(P<0.05);After giving Xiao Fu Tong Jie Fang each dose group and positive drug group,AVP and c AMPexpression levels reduced compared with the model group(P<0.05).(2)The influence of AQP3:Compared with the blank group,model group,AQP3 were the most strongly expressed both in cell membrane and kytoplasm.After giving Xiao Fu Tong Jie Fang each dose group and positive drug group,AQP3 were more less strongly expressed both in cell membrane and kytoplasm;(3)The influence of AQP1 m RNA?AQP2m RNA?AQP3m RNA inkindney and colon:Compared with the blank group,model group,Levels of AQP1 m RNA?AQP2m RNA?AQP3m RNArelative expression levels were significantly increased(P<0.01);After giving Xiao Fu Tong Jie Fang each dose group and positive drug group,AQP1 m RNA?AQP2m RNA?AQP3m RNAexpression levels reduced compared with the model group(P<0.01).(4)The influence of AQP1?AQP2 and AQP3 inkindney and colon:Compared with the blank group,model group,Levels of AQP1?AQP2 and AQP3 relative expression levels were significantly increased(P<0.01);After giving Xiao Fu Tong Jie Fang each dose group and positive drug group,AQP1?AQP2 and AQP3 expression levels reduced compared with the model group.Conclusions:1.Xiao Fu Tong Jie Fang can improve the general condition of each group.The water content of the colon were significantly increased;2.The kindey can affect water metabolism disorder in colon by adjusting ALD's synthesis and secretion.This may be one of the scientific connotation of nursing functional constipation from the kindey;Xiao Fu Tong Jie Fang can affect water metabolism disorder in colon by adjusting ALD's synthesis and secretion to nursing functional constipation;3.The water metabolism disorder in kidney is closely related to the functional constipation.The kindey can affect water metabolism disorder in colon by adjusting aquaporins.This may be one of the scientific connotation of nursing functional constipation based on the kindney controlling water metabolism and governing;Xiao Fu Tong Jie Fang may affects the aquaporin water channels in the kidney to regulating body's water metabolism and correcting colon's water metabolism disorder to nursing the functional constipation.
