Objective: to campare the serum protein levels of Han and Uygur patients with locally advanced or advanced esophageal squamous cell carcinoma before and after radiotherapy and chemotherapy,to investigate the expression characteristics of serum differential proteins in the two ethnic groups,and to predict the difference of serum proteins in the short-term and long-term efficacy of chemoradiotherapy and analyse pathway.Method: 1,collected 35 han patients and 28 uygur patients with locally advanced or advanced ESCC,and blood samples before and after radiochemotherapy,at the same time to collect normal people blood as a control,evaluate curative effect and follow-up.2,each group serum using i TRAQ technology peptides tag,of the ms spectra by using the PD software filtering,after extraction of the spectra with mascot for retrieval,and through the PD software for further quantitative analysis,by using R software to screen different proteins according to different groups,search the relevant information of the difference or target protein retrieval function in database,and carry out the biological function enrichment analysis.Results: 1.different ethnic have different characteristcs in serum protein expression:(1)a total of 46 proteins different expression significantly in the han patients before radiochemotherapy group compared with normal control group;after radiochemotherapy,a total of 22 proteins different expression significantly;21 proteins different expression between after and before group.Through screening and verification in database,choose the difference ratio 1.5 to 2 times,the significant and meaningful protein such as platelet factor 4(PLF4),galactose binding lectin binding protein 3(LG3BP),24(SPP24)secreted phosphoprotein,zinc-alpha-2-glycoprotein(ZA2G),silk protein(COF1),C-reactive protein(CRP);(2)a total of 105 proteins different expression significantly in the uygur patients before radiochemotherapy group compared with normal control group;after radiochemotherapy,a total of 114 proteins different expression significantly;24 proteins different expression between after and before group.Through screening and verification in database,choose the difference ratio 1.5 to 2 times,the significant and meaningful protein such as transferrin variant,stimulating peptide 1(KNG1),-alpha trypsin inhibitor heavy chain between H1 and H2,serum amyloid A(SAA),set of protein and insulin-like growth factor(insulin-like growth factor,IGF),rich in histidine glycoprotein(HRG),lipopolysaccharide binding protein(LBP),glyceraldehyde 3-phosphate dehydrogenase(GAPDH).2.Expression characteristics of differentially expressed proteins in different ethnic groups in predicting curative effect of chemoradiotherapy:(1)a total of 17 proteins different expression significantly in the han patients after radiation and chemotherapy between the recent curative effect is invalid group and effective group in the recent curative effect;there were 15 patients with esophageal cancer in OS 1~2 years group,and 12 patients in OS 2~3 years of esophageal cancer,10 differentially expressed proteins were screened,and all of them were down-regulated proteins.There were 15 patients with esophageal cancer in OS 1~2 years group,and 8 patients with esophageal cancer in OS>3 years group.10 differentially expressed proteins were screened.Through the screening and verification of the database,choose the difference ratio 1.5 to 2 times,significant and meaningful protein such as polymer immunoglobulin receptors(PIGR),adipose cytokines,serine protease inhibitors,alpha acid glycoprotein,haptoglobin.(2)A total of 75 proteins different expression significantly in the uygur patients after radiation and chemotherapy between the recent curative effect is invalid group and effective group in the recent curative effect;there were 16 patients with esophageal cancer whose PFS was more than 1 year,there were 12 patients with esophageal cancer whose PFS was less than 1 year,comparing the two groups,there were 153 different proteins were screened;through the screening and verification of the database,choose the difference ratio 1.5 to 2 times,the significant and meaningful protein such as serum amyloid protein,immunoglobulin,retinol binding protein(RBP)and extracellular superoxide disproportionation enzyme,fibronectin,agglutination in protein 1;3.Analysis of KEGG pathway of serum differential protein in han and uygur pantients with esophageal squamous cell carcinoma:(1)the different proteins of han pantients with esophageal squamous cell carcinoma before and after treatment were imported into KEGG database,there were three pathways wich were P<0.2 and related to tumor,such as small cell lung cancer pathway,chemokine signaling pathways,thyroid carcinoma;(2)the different proteins of uygur pantients with esophageal squamous cell carcinoma before and after treatment were imported into KEGG database,there were two pathways wich were P<0.05 and related to tumor,such as the Toll-like receptors(TLR)pathway,NF-k B signaling pathways;(3)after introducing the differentially expressed proteins related to the efficacy of Han patients with esophageal cancer into the KEGG pathway,one of the tumor-associated pathways was screened in all signal pathways,which is the HIF-1 signaling pathway;(4)the different proteins correlated with treatment effect of uygur pantients with esophageal squamous cell carcinoma were imported into KEGG pathway,and there was one pathway(P<0.05)in the KEGG database with statistical difference,and PPAR signaling pathway was the tumor related pathway.Conclusion:(1)through the screening of serum proteins from peripheral blood of patients of Han and Uygur ethnic groups with esophageal cancer,serum proteins that are valuable for diagnosis have been screened.Some proteins have confirmed the results of previous studies.Some proteins are first discovered and may have potential diagnostic value,such as blood conjugates.(HEMO),haptoglobin(HPT),SPP24,etc;(2)compared with the serum proteins of short-term efficacy and long-term efficacy between han and uygur patients with esophageal cancer,we have screened some serum proteins that were meaningful in the evaluation of efficacy.(3)compared the different proteins between the two ethnic groups,some proteins of diagnosis and curative effect judgment are similar,some are different,such as a-acid glycoprotein(A1AG)and haptoglobin(HPT),histone,are prognosis protein in both ethnic groups;(4)the activation of TLR and nf-kb signaling pathway indicated poor prognosis,while the chemokine pathway suggested good prognostic factors.PPAR,hif-1,VEGF,Wnt pathway have also been identified as specific pathways of the tumor,which can be used as the focus of future research.
