Objective:To observe the clinical effect of Jin Que Liang Xue Particle(JQLX)in the treatment of eczema.To explore the possible mechanism of JQLX by using rat eczema model lesion,the difference of swelling degree and sera of IL-2,IL-4,IL-1? TNF-?,IFN-y and IgA,IgG,IgM,C3,C4 levels,spleen index and thymus index,IL-1? TNF-a mRNAs gene and P-P38,NF-K B65 proteins expreesion.Methods:To observe the morphological changes of skin lesions in mice with eczema.The model of eczema was induced by 2,4-dinitrochlorobenzene(Mock),for High/Medium/Low-JQLX drug intervention,were treated with JQLX control group,Compound Glycyrrhizin positive control group(CG)and blank Group(NC).According to Zhao Ban EASI score method to score to evaluate the efficacy of erythema,edema,exudation,epidermal exfoliation and other skin lesions changes.The levels of IL-2,IL-4,IL-1?,TNF-?,IFN-?,IgA,IgG,IgM,C3,C4 in the serum of the eczema model were determined by ELISA.Score method to score to evaluate the spleen index and thymus index of NC,Mock and High/Medium/Low-JQLX.RT-PCR was used to detect the content of IL-1?TNF-a mRNA in the eczema model of NC,Mock and High/Medium/Low-JQLX.Western Blot method was used to detect the expression of P-P38,NF-kB65 protein in the patients with NC,Mock and High/Medium/Low-JQLX.Results:JQLX group was used to control the eczema model of rat.Compared with CG and model group,there were sigificant differences in erythema,papules,exudation and epidemis exfoliation(P<0.05),and the difference between the two goups was significantly higher than that of the model group(P<0.05).The efficacy of the index was significantly better than that of CG.Levels of IL-2,IL-4,IL-1? TNF-?,IFN-?,IgA,IgG,IgM,C3,C4:There were significant differences in IL-2,IL-4,IL-1?,TNF-?,IFN-?,IgA,IgG,IgM,C3,C4 levels in the group with Medium-JQLX,CG,Mock and NC.The level of IL-2 and IFN-? in the Mock group significantly reduced.lower than that in the NC group(P<0.05),The level of IFN-? in the Mock group significantly reduced.Lower than that in the NC group(P<0.05).The levels of IL-4?IL-1? and TNF-? in the Mock group significantly inereased,higher than that in the NC group(P<0.05),The level of IL-2 and IFN-? in the Medium-JQLX group was significantly inereased(P<0.05).Higher than that in the Mock group(P<0.05).The levels of IL-4,TNF-? and IL-1? in the Medium-JQLX group was significantly inereased lower than that of the Mock group.The indexs of spleen and thymus the Mock group significantly reduced.lower than that in the NC group(P<0.05).Compared with Medium-JQLX and Mock group,there were sigificant differences in spleen index and thymus index(P<0.05),and the difference be tween the two goups was significantly higher than that of the Mock group.The index of thymus the Mock group and High/Medium-JQLX significantly differences inereased.There was a significant difference(P<0.05).The results showed that the Medium-JQLX group could significantly increase the spleen index of the CG group.The High/Medium-JQLX group could significantly increase the thymus index and improve the immune organ function of eczema rats,which was better than the CG group.The levels of IgA?IgM?IgG in the Mock group significantly inereased:compared with the NC group,the serum levels of IgA,IgM and IgG in the Mock group were significantly increased,with statistical difference(P<0.05),suggesting that the immune function of eczema rats was unbalance.After the treatment,the levels of IgA,IgM and IgG in the serum decreased significantly after the treatment of the Medium-JQLX group.There was a significant difference in the model group(P<0.05),and the middle dose effect was better than that of the CG group.The results showed that Medium-JQLX reduce the content of immunoglobulin IgA,IgM and IgG in humoral immunity of rat eczema,and the effect of regulating humoral immunity was better than that of CG group.The levels of C3 and C4 in the Mock group significantly inereased.Higher than that in the NC group(P<0.05),suggesting that the immune function of eczema rats was unbalance.After the treatment,the levels of C3 and C4 in the serum decreased significantly after the treatment of the Medium-JQLX group.There was a significant difference in the model group(P<0.05),and the middle dose effect was better than that of the CG group.The results showed that Medium-JQLX reduce the content of immunoglobulin C3 and C4 in humoral immunity of rat eczema,and the effect of regulating humoral immunity was better than that of CG group.The efficacy of the index was significantly better than that of CG.IL-1?,TNF-? mRNAs and P-P38,NF-?B65 proteins There was significant difference between the two groups(P<0.01),There were significant differences between the two groups(P<0.01).Compared with CG group,the patients were treated with JQLX group and CG group,the difference was statistically significant.Conclusion:The clinical effect of JQLX is siginificant,which can eliminate the effect of erythema,blisters,pimples and reduce oozing.The basic experiment can reduce the degree of skin lesion swelling,reduce erythema,Epidermal exfoliation and other inflammatory response,the effective regulation of skin lesions in the IgA,IgG,IgM,C3,C4 spleen index and thymus index.JQLX can blunt inflammation by inhibiting the TNF-? and IL-1? mRNAs and P-P38 and NF-kB65 proteins leves of the P38MAPK/NF-?B pathway.It suggests that the JQLX particle may reduce the inflammation reaction in the rat eczema model by inhibiting the activation of the P38MAPK/NF-?B pathway.
