ANK is a new oncogene encoded protein found in tumor cells in recent years.It has been widely concerned because of its overexpression in malignant tumor tissues.It is thought that ANK2 gene is closely related to many oncogenes and tumor suppressor genes,which leads to the occurrence and development of tumor cells.The accurate examination of the value of ANK2 can not only provide the clinicians with the patient's information,but also provide a new and effective method for the detection of the disease.Therefore,further study of the effect and molecular mechanism of ANK2 on hepatocellular carcinoma is of great significance for understanding the pathogenesis and progression mechanism of primary liver cancer.This study detected the difference in expression of ANK2 in primary liver cancer and adjacent cancer tissue,which laid the foundation for further study of the function of ANK.The expression of ANK2 gene in hepatocellular carcinoma is demonstrated by experiments.This experiment illustrates the effect of ANK2 on the prognosis of primary liver cancer patients and the survival period after the operation.It provides a new theoretical and reference basis for the pathogenesis and targeted therapy of hepatocellular carcinoma in the future.Objective To detect the difference of ANK2 gene expression in primary hepatic carcinoma and related adjacent tissues,and to analyze the its effect on prognosis of liver cancer patients.Methods Surgical specimens of 80 primary hepatic carcinoma patients were collected.Quantitative polymerase chain reaction(q-PCR)and immunohistochemistry were used to detect the expression of ANK2 gene in cancer and adjacent tissues.According to the expression level of ANK2 gene in primary hepatic carcinoma,the patients were divided into high expression group(n=40)and low expression group(n=40).The sex,age,AFP level,Child-pugh score were recorded and included in the experiment.Univariate and multivariate analyses were used to analyze the prognosis of patients based on COX regression model.Results In 80 primary hepatic carcinoma samples,nearly 70%of the(55/80)tumor tissue samples showed dysregulated expression of ANK2 and were significantly higher than those in the adjacent tissues.After grouping patients according to ANK2 expression levels,it was found that the overall survival rate(P=0.024)and disease-free survival rate(P=0.011)of the patients with low expression of ANK2 were significantly higher than those of the patients with high expression group.COX univariate and multivariate analysis showed that ANK2 expression level was an independent prognostic factor.Conclusion The expression of ANK2 gene in primary hepatic carcinoma tissues is abnormally higher,and affects the prognosis of patients.
