Background of research:Acute pancreatitis(AP)is the most common alimentary system disease.Its pattern of manifestation is the self-alimentary of trypsase,which has mild condition and the trait of self-limitations.However,20%to 30%AP patients may develop into severe acute pancreatitis(SAP),and the overall mortality is 5%to 10%.Therefore,the key point of AP clinical diagnosis and treatment is exploring the worsening mechanism of pancreatitis and seaching possible intervention methods.the complex nosogenesis,long course of disease,long-term diet forbidden and many complication of AP will impair the intestinal barrier function,disturb intestinal bacteria colonies and change the position of bacteria.For this reason,the intestinal microecological balance has great effect on trasforming and returning AP.However,the intestinal microflora composition of AP is unclear.In this way,identifying the trait of intestinal bacteria colony is of great importance.This research is based on high-flux 16SrRNA's metagenomic sequencing technology,to construct the constitute of intestinal microecological core flora genera of AP patients and study systematically the relative between AP and intestinal core bacteria micoflora,which providing reliable theory support for future AP microecological treatment.Aims:compare the difference of intestinal microecological core bacteria flora genera between AP patients and healthy subjects by intestinal microecological core bacteria flora genera's metagenomic sequencing technology.Methods:(1)Collecting the fecal sample of 48 AP patients and 50 healthy subjects;appraising the genera type of subjests'DNA in stools bacteria flora through high-flux 16SrRNA genome testing system;polishing up the quality of the results,in order to construct the testing system of intestinal microflora.(2)In terms of identifying clearly the microflora,analyzing the different traits of bacteria flora in stools between AP patients and healthy subjects through non-limited sequence method PCoA?Results:(1)Comparing with normal control,the AP patients have higher lever in total number of white blood cell,rate of NE,glu,glutamic-Ocala Eric transminase,glutamic-pyruvic transminase,total bilirubin,CA199.While they have lower level in hemoglobin,hematocrit and total cholesterol.(2)There is distinctive difference of genera level effective sequence distribution between AP patients and normal control group,especially in the aspects of the total number in microbe genera and its diversity.The proportion of PoCA PC1,PC2,PC3 in two groups is respectively 43.15%,13.55%and 6.99%,sugessting the difference of microflora in two groups.(3)The cluster analysis of UPGMA indicated that there were totally 27 different types of microbe core flora in two groups.AP intestinal bacteria flora mainly composed of bacteroidetes and proteobacteria,while the healthy control group mainly consist of poster wall of bacteria and actionobacteria.Conclusion:(1)High-flux 16SrRNA sequence and analysis can be taken as a reliable appraisal system to identify intestinal microflora genera.(2)AP patients has distinct difference in intestinal microbe core flora and genera comparing with the healthy subjects.It can be a new treatment of AP to intervention accurately intestinal microbe genera.
