percent are CAG,CSG may develop into CAG.if persistent refractory,so,it is very important to prevent it from developing CAG and gastric cancer.now,Western medicine is only symptomatic treatment,the method is not only unsatisfactory but also easy recidivation.On the contrary,traditional Chinese medicine has special superiority and potential in discriminate treat?improving symptom and preventing recidivation Anwei pills were good prescription that summed up clinical practice in several decades.Clinieal application winned award-good effect..So,it deserved the further experimental research.Objective:To establish a rat model of chronic gastritis,to probe into Effect of anwei pill on inflammatory cytokines in blood serum and expression of cell proliferation controlling protein?NF-?B p65 mRNA in gastric mucosa of CG rats in level of protein and gene,in order to further provide its mechanisms.Method:Animal model of SD rats with CG was established by combined administration of 60%ethanol,20mmol·L-1sodium deoxycholate and 0.05%ammonia water and irregular diet for 13 weeks,anwei pills used to treat CG rat for 30 days.Pathologic changes of the gastric mucosa were studied by microscope,Expressions of cell proliferation related controlling protein(PCNA)and(EGFR)from gastric mucosa of CG model rats were measured by immunohistochemistry and the changes of them were also analyzed by image analysis;the expression of NF-?B p65 mRNA in gastric mucosa of CG rats were detected in situ hybridization;Changes of blood serum IL-2?IL-4?TNF-??Gas?somatostatin and EGF in CG rats were measured by enzyme-linked immunosorbent assay(ELISA);levels of total acids,free acids,PePsase activity and free mucus in gastric juice of gastritis rats were measured with instrument.To establish a Balb/c mouse model with Hp infectious gastritis through administration of HP.anwei pill was adminidtrated for 20 days,Hp was detected by rapid urease test,Giemsa dying,and bacterial culture,and histopathologic changes in the gastnc mucosa of mice were determined by HP staining.Result:1.60%alcohol and 20mmol.L sodium deoxycholate/0.05%ammonia water and irregular diet were given to rats for 12 weeks,inflammation grades of gastric antrum of model group were less than those of the model group(P<0.01),the means of ratios of the thickness of gastric mucosal gland and muscularis mucos(L1/L2)were very close to normal group(P>0.05).Pathologie result show the rats model of CG is convincing.2.the slight change of pathology and a little inflammatory infiltration were observed in anwei pill and TCM control groups,inflammation index of gastric antrum were less than those of the model group(P<0.05),the means of ratios of the thickness of gastric mucosal gland and muscularis mucos(L1/L2)in all groups were very close to those of model group(P>0.05).3.results measured by immunohistochemistry show that Compared with the control group,Protein expressions of PCNAand EGFR in gastric mucosa of CG model rats raised significantly.anwei pill and TCM control group might decreased expressions of PCNA and EGFR in some degree,the effect of anwei pill in medial dose were very close to that of TCM control.groups4.results detected in situ hybridization show that expressions of NF-?B p65 mRNA in gastric mucosa cells of CG model rats raised significantly and irregularly,and optical density and a rate of positive square measu increased significantly,anwei pill and TCM control might decreased expressions of NF-?B p65 mRNA,optical density and a rate of positive square measue of NF-?B p65 mRNA in anwei pill and TCM control groups.were lower than those of model5.results detected by ELISA show that EGF,GAS,IL-2 and TNF-ain model groups were higher than those in normal control group.IL-4 was lower than those in normal control group,Somatostatin were very close to normal model group(P>0.05),anwei pills in high and medial dose,TCM control decreased the concentration of gastrin and TNF-a,increased the level of IL-4,anwei pills in medial dose decreased the concentration of EGF,increased the level of IL-2,Somatostatin in all groups had few change.6.the concentration of free acid and total acid were significant increase,free mucin contents and PePsase activity were significant higher in model group,anwei pills and TCM control reduced the concentration of free acid and total acid,PePsase activity,anwei pills in high and medial dose reduced free mucin contents,TCM control had little effects on it.Conclusion:1?Anwei pills in medial and high dose could relieved inflammation of gastric mucosa in CG rats,no effect of Anwei pills on the thickness of gastric mucosal gland/mucosal muscularis.2?the concentration of freeacid,total acid and PePsase activity were significant decrease in anwei pills group,Anwei pills in low dose had not effect on content of free mucin3?Anwei pills can restrain the excessive expression of EGFR?PCNA in gastric mucosa of CG.rats which may one of mechanism that treat chronic gastritis(P>0.05)?4?Anwei pills could obviously decrease positive cell of expression of NF-?B p65 mRNA in gastric mucosa.The optical density value and rate of positive area of NF-kB mRNA were reduced obviously.5?Anwei pill could restrain the secretion of Gastrin and epidermal growth factor,reduce concentration of TNF-?and IL-2,raise IL-4 in blood serum.no Effect of anwei pill on somatostatin in CG rats. |
