The search about structure and reaction of proteins and small drug-related molecules under physiological conditions is of great significance for understanding the corresponding physiological effects with relevant proteins of drugs in the body.One-dimensional?1D?and two-dimensional?2D?infrared pump probe system can provide structure and dynamic information for the sample molecular effectively.Therefore,the existing 1D UV pump IR probe system was used to study tryptophan in aqueous solution which is a typical fluorescent probe,2D IR pump probe system was constructed and the drug of carbon monoxide release molecules?CORMs?and water high pressure were also studied by the system further.This thesis mainly includes the following four achievements:?1?The photochemistry and photophysical processes of tryptophan aqueous solution excited by 266 nm light were explored by using the UV pump IR probe system.We obtained the vibrational information of Trp-+ generated during the photoionization process and the excited singlet state Trp1* generated during Triplet particle generation process.It was found that the generation time of Trp-+ and Trp1* is about 800 fs and 10 ps respectively,and the former process is connected with the electron transfer.?2?2D IR system was built up.IR light is generated by commercial optical parametric amplifier?OPA?and non-linear optical differential generator?NDFG?.The system adopts pump probe configuration,in which two pump beams with time interval of ? are generated by pulse shaper.Due to the high cost of multi-channel detectors of mid-IR spectrum,the probe light is converted to visible light by sum frequency generation with 800 nm pulses which can be detected by cheap visible light array camera.?3?2D IR system was used to explore the reaction kinetics of CORMs and the interaction process with proteins.CORM-2 has been proved to have effective physiological therapeutic effect.The reaction process of CORM-2 and its solvent DMSO within 018h was obtained by 2D IR spectroscopy.CORM-3 is the first water-soluble CORMs that had been verified with effective physiological therapeutic effect,so we studied the reaction between CORM-3 and water,further the interaction process with lysozyme.?4?2D IR system was used to study the structural changes of hydrogen bond network in aqueous solution under high pressure.It was concluded that there are liquid-liquid phase transition point under pressure around 0.8 GPa.
