The Influence Of Huangqibaoxin Decoction And The Effective Components Salvianolic Acid B On Myocardial Fibrosis And TGF-?/smad Singnal Of Rats With Dilated Cardiomyopathy

Objective:1.The present study investigate the effect of Huangqiaoxin Decoction on myocardial fibrosis of rats with Dilated cardiomyopathy and clarify the possible mechanism on TGF-?/smad singnal pathways.2.The present study investigate the effect of salvianolic acid B-an effective composition ofHuangqiaoxin decoction on cardiac fibroblasts proliferation and collagen and explore the possible mechanism on TGF-?/smad singnal pathways.Methods:1.Dilated myocardiopathy were induced by doxorubicin injected intraperitoneally for 6 weeks.Rats that had demonstrated dilated myocardiopathy were randomly divided into five groups except control group.Three groups were treated with BXD(7.5/kg,15g/kg,30 g/kg)daily for 4 weeks.One group was treated with 8.75g/kg of Captopril(positive control),and with physiologic saline(negative control).Cardiac function was evaluated by echocardiography.Hematoxylin and eosin(HE)and Masson's trichrome staining were performed,PICP and P?NP were assessed by ELISA,and the expression of galectin-3 and collagen-type ? and ? were evaluated with real-time reverse transcription-PCR.and interrelated proteins TGF-??smad were detected by western blot.2.Cardiac fibroblasts were cultured in vitro and stimulated with 10-7 mol/L Angiotensin,then interfered with salvianolic acid B at difference concentrations(10-5 mol/L.10-6 mol1/L and 10-7 mol/L)and valsartan.Cardiac fibroblasts proliferation assessed by CCK8,Galectin-3 were assessed by immunofluorescence,and the expression of collagen-type ? and ? were evaluated with real-time reverse transcription-PCR.and interrelated proteins on interrelated proteins TGF-??smad were detected by western blot.Results:1.Huangqibaoxin decoction down-regulated Gal-3,C.ol ?,and Col ? and correlate with expression levels of fibrosis markers.It also significantly decreased myocardial collagen volume fraction,together with markedly prevented the upregulations of Collagen ? and Collagen ?.Moreover,BXD downregulated expressions of TGF-b1,Smad3 in the myocardial fibrosis rats.2.Salvianolic acid B down-regulated Gal-3,Col ?,and Col ? mRNA.It also significantly prevented the upregulations of Collagen ? and Collagen ?.Moreover,Salvianolic acid B downregulated expressions of TGF-b1,Smad3 in the Cardiac fibroblasts.Conclusions:Huang,qibaoxin decoction can improve heart function and relieve ventricular remodeling in rats with DCM.
