Objective: pulmonary fibrosis is the most common interstitial lung disease, which was often classified as feibi in traditional Chinese medicine. Study show that some Chinese herbal medicine in the treatment of pulmonary fibrosis is closely attributed to their antioxidant effect. This subject belongs to the sub-topic of National Natural Science Foundation "Eliminating Phlegm Traditional Chinese Medicine Regulate Antioxidant Mechanism in Pulmonary Fibrosis Through Multiple Pathways(NO.81273704). " Several group members collaborate on this subject.Therefore, in this study, we selected out eight eliminating expectorant herbs to treat on bleomycin induced pulmonary fibrosis rat model and observed antioxidant index, to investigate the antioxidation mechanism of eliminating expectorant herbs in pulmonary fibrosis in.Method: A total of 110 male SPF Wistar rats were randomly divided into: eight Chinese medicine group( Arisaema erubescens Schott,Fritillaria cirrhosa, Peucedanum declursivum Maxim, Sterculia lychnophora Hance Pinellia ternata Breit, Cynanchum glaucescens, Airpotato Yam Rhizome and Peucedanum praeruptorum, namely Group A-H),Pirfenidone group( group X), control group( group Y), Blank control group(group Z),10 rats in each group. By intratracheal rapid drop injection of bleomycin solution method established a rat model of pulmonary fibrosis. 3 days after modeling, the rats in the experimental group were respectively given corresponding Chinese herb medicine(Group A-H), pirfenidone( group X) and physiological saline( group Y) by gavage treatment and executed after 28 days. Then took out the intact lung tissue to observe the appearance and pathological section and to measure the content of SOD, MDA, GSH, Nrf2 in lung tissue homogenate through ELISA method and chemistry fluorescence test.Result:1. Appearance of lung tissue: lung tissue of rats in all the experimental groupsappeared different degree of flaky ecchymosis and hemorrhage, increased hardness of the lung lobes, which was accompanied by diminished pulmonary parenchymal, showing grayish white, uneven surface with nodular change.2.Pathological section observation: the lung tissue of all the experimental groups of rats under light microscope showed different degrees of interstitial capillary congestion,alveolar septal thickening, inflammatory cell infiltration and fibrous tissue proliferation.3.(1)Changes in the content of SOD in lung tissues of rats: the content of SOD in lung tissue of blank control group( group Z) was higher than control group( group Y)(P <0.01); group B?G?X?E?F were significantly increased compared to group Y(P < 0.05);group B?G?E?F and group X showed no difference(P > 0.05).(2) Changes in the contents of MDA in lung tissues of rats: in group Y, the content of MDA was more than group Z(P< 0.01).B?C?E?G?X were significantly decreased compared to group Y(P < 0.05); and MDA was lower in group B than in group X decreased significantly(P < 0.05).(3) The content of GSH in lung tissue of rats: in group Y, GSH expression was significantly lower than in group Z(P < 0.01). GSH content in group B?D?E?X was significantly higher than in group Y(P < 0.01), and it was higher in group E than in group X(P < 0.01).(4) Changes in the content of Nrf2 in the lung tissue of rats: Nrf2 expression was higher in group Y compared with group Z(P < 0.01). The content of Nrf2 increased significantly in group B?C?D?E?H?X compared to group Y,(P < 0.05); and there was no remarkable difference between group B?C?D?E?H and group X(P > 0.05).Conclusion:1. This experiment again confirms the reliability and stability of pulmonary fibrosis rats model induced by intratracheal instillation of bleomycin of.2. Fritillaria cirrhosa, Peucedanum declursivum Maxim, Cynanchum glaucescens and Peucedanum praeruptorum had an equivalent effect in increasing the activity of SOD,clearing oxygen free radical in the enzyme catalytic antioxidant pathways compared with pirfenidone; Fritillaria cirrhosa showed a better effect in improving antioxidant indexes of MDA and inhibition of lipid peroxidation products generated in the way of antioxidant than that of pirfenidone; Peucedanum declursivum Maxim in GSH directly or by indirect enzyme catalytic reduction oxidation of hydrogen peroxide and lipid peroxide reduction pathway function is better than that of pirfenidone; Fritillaria cirrhosa, Pinellia ternata Breit, Arisaema erubescens Schott, Peucedanum declursivum Maxim and Sterculialychnophora Hance these five Chinese medicine act equally in improving the Nrf2 antioxidant index and the regulation of antioxidant gene expression compared with pirfenidone group.3. Through this study, we can provide a theoretical basis for clinical application of traditional Chinese medicine in treatment of pulmonary fibrosis which can further guide the clinical treatment.
