Objective:1.To summarize characteristics distribution of Chinese medicine syndromes in low endometrial receptivity patients.This study aimed to provide an experimental basis to clinical research and tried to provides new evidences to conducive to embryo implantation.2.To investigate the effects of Erbuzhuyu decoction in SCFmRNA expression,HOXA-lOmRNA and other related regulations' expression.To demonstrate new targets and regulatory mechanisms of Erbuzhuyu decoction improving endometrial receptivity,which also can raise up success rate of IVF-ET.Method:Clinical research:Firstly,designed low endometrial receptivity clinical questionaire according to literature review.104 patients which matched inclusion criteria of low endometrial receptivity were collected on the method of clinical observation.Then analyze and classify Chinese medicine syndromes types of those 104 patients and used SPSS17.0 for statistical processing.Finally,summarized distribution characteristics of Chinese medicine syndromes in low endometrial receptivity patients.Experimental research:1.Rats were subjected to embryo implantation dysfunction model in GnRHa+HMG+HCG method.This model was considered the closest model to clinical patients who were accepted the IVF-ET therapy.2.Real-time PCR was used to test the expression of SCFmRNA and HOXA-lOmRNA under the influence of different doses of Erbuzhuyu decoction,Progynova and Aspirinin.The study were mainly objected to explore the regulation changes on the expression of SCFmRNA and HOXA-lOmRNA in model endometrium.3.Immunohistochemistry was used to test the expression of endometrial LIF,ER,PR,???3.Via observing the expression of those related regulation,we try to figure out the influence of Erbuzhuyu decoction on endometrium receptivity.Results1.Among those common symptoms and signs of 104 patients,a total of 14 symptoms appeared frequently(more than 30%).There were of the sequence from high to low as follow:hypomennorrhea,soreness and weakness in the lumbar region and legs,dizziness,tinnitus,or frequent night urination,thin coating, hyposexuality,bleak blood,aversion to cold,deep and thin pulse,dark red tongue,petechiae,purple dark blood,clots,early menstrual period,dysmenorrhea,thereamong,dizziness,tinnitus,dark and pale tongue,clear and long urination,clear urine long,soreness and weakness in the lumbar region and legs,hypomennorrhea,referred to kidney-Qi deficiency.Purple dark blood,clots,dysmenorrhea,dark red tongue,petechiae,those symptoms and signs above referred to blood stasis.2.Through cluster analysis,the main pathogenesis of low endometrial receptivity patients referred to kidney-Qi deficiency with blood stasis symptoms and signs.3.The expression of SCFmRNA of the model group was significantly lower than the control group(P<0.05).Compared with the model group,the expression of all the positive groups and the Erbuzhuyu groups were significantly increased(P<0.05),showed that the positive samples and the Erbuzhuyu decoction can promote the expression of SCFmRNA;The middle-dose Erbuzhuyu group and high-dose Erbuzhuyu group had more obvious effects than low-dose Erbuzhuyu group(P<0.05).The expression of SCFmRNA of middle-dose Erbuzhuyu group and high-dose Erbuzhuyu group had no difference(P>0.05),but both were higher than Progynova group(P<0.05).The expression of SCFmRNA of each Erbuzhuyu group was significantly higher than Aspirin group(P<0.05).4.The expression of HOXA-lOmRNA of the model group was significantly lower than the control group(P<0.05).Compared with the model group,the expression of all the positive groups and the Erbuzhuyu groups were significantly increased(P<0.05),showed that the positive samples and the Erbuzhuyu decoction can promote the expression of HOXA-lOmRNA;The expression of HOXA-lOmRNA of middle-dose Erbuzhuyu group and low-dose Erbuzhuyu group were significantly higher than Progynova group and Aspirin group(P<0.05).5.The expression of ? ? ? 3 of the model group was significantly lower than the control group(P<0.05).Compared with the model group,the expression of all Erbuzhuyu groups were significantly increased(P<0.05),showed that each doses of Erbuzhuyu decoction can promote the expression of ? ? ? 3;The expression of ? ? ? 3 of all Erbuzhuyu groups were significantly higher than Aspirin group and Progynova group(P<0.05).6.The expression of LIF of the model group was significantly lower than the control group(P<0.05).Compared with the model group,the expression of middle-dose Erbuzhuyu group,low-dose Erbuzhuyu group and Aspirin group were significantly increased(P<0.05),showed that middle-dose Erbuzhuyu group,low-dose Erbuzhuyu group and Aspirin group can promote the expression of LIF;The expression of LIF of middle-dose Erbuzhuyu group and low-dose Erbuzhuyu group were were significantly higher than Aspirin group(P<0.05).The expression of LIF of low-dose Erbuzhuyu group was significantly higher thanProgynova group(P<0.05).7.The expression of ER of the model group was significantly lower than the control group(P<0.05).Compared with the model group,the expression of middle-dose Erbuzhuyu group and low-dose Erbuzhuyu group were significantly increased(P<0.05),showed that middle-dose and low-dose Erbuzhuyu decoction can promote the expression of ER.The expression of ER of middle-dose Erbuzhuyu group and low-dose Erbuzhuyu group were significantly higher than Progynova group and Aspirin group(P<0.05).8.The expression of PR of the model group was significantly lower than the control group(P<0.05).Compared with the model group,the expression of each Erbuzhuyu groups was significantly increased(P<0.05),showed that all doses of Erbuzhuyu decoction can promote the expression of PR.Conclusion1.The main pathogenesis of low endometrial receptivity patients referred to kidney-Qi deficiency with blood stasis.There were syndromes and signs of kidney-Qi deficiency with blood stasis.2.Erbuzhuyu decoction can significantly upregulate the expression of endometrial ER,PR,LIF,? ? ? 3,SCFmRNA and HOXA-lOmRNA,struggesting that Erbuzhuyu decoction had effective influence on improving endometrium receptivity.It had a multi-target,and multi-link mechanism.
