Purpose:1.to research effects and mechanisms of Erbuzhuyu improving prescription on the endometrium of mice model with dysfunctional embryo implantation.2.to observe effects of Rhizoma drynariae and its main components on endometrial receptivity in a mouse model of dysfunctional embryo implantation,and search the active constituents preliminarily.3.to explore effects and mechanisms of Erbuzhuyu improving prescription on endometrium receptivity of patients with repeated IVF-ET failure,and contribute on the elevation of pregnancy rate in clinical application.Method:Experimental research:Rats were subjected to embryo implantation dysfunction model in GnRHa+HMG+HCG method.1.SEM was used to research effects on morphology of pinopodes in mice endometrium of groups of Erbuzhuyu Decoction,Erbuzhuyu improving prescription,progynova and aspirin,the same as Rhizoma drynariae,total flavone and naringin.2.qPCR was used to research effects on the mRNA expressions of SCF,HOXA10 in mice endometrium of groups of Erbuzhuyu Decoction,Erbuzhuyu improving prescription,progynova and aspirin,the same as Rhizoma drynariae,total flavone and naringin.3.Immunohistochemistry was used to research effects on the expressions of ER,PR,LIF,and integrin ?v?3 in mice endometrium of groups of Erbuzhuyu Decoction,Erbuzhuyu improving prescription,progynova and aspirin,the same as Rhizoma drynariae,total flavone and naringin.Clinical research:Collected data of patients with repeated IVF-ET failure,After three months of Erbuzhuyu improving prescription intervention,collected mid luteal phase endometrium by endometrial biopsy.HE staining and electron microscopy were used to study the morphology of pinopodes.qPCR was used to research the mRNA expressions of SCF,HOXA10,MMP-9.Immunohistochemistry was used to measure the effect of molecular biology,such as LIF and integrin ?v?3.Results:1.In the study on the effect of Erbuzhuyu improving prescription in endometrium of mice model with dysfunctional embryo implantation,the expression of each index model group was lower than that of the blank group(P<0.05).Compared with the model group,the endometrium of Erbuzhuyu improving prescription group was more regular and the expression of pinopodes was abundant,and the gap between the cells was obvious,which is good for cell adhesion;the Erbuzhuyu improving prescription group had an obviously higher LIF,ER and PR expression in mice endometrium,compare with the western medicine groups,Erbuzhuyu improving prescription had similar level(P>0.05);The Erbuzhuyu improving prescription had an obviously higher ER expression compared with the progynova group(P<0.05),similar to the aspirin group(P>0.05);the Erbuzhuyu improving prescription had an obviously higher PR,LIF,?v?3 integrin expression,had a similar level compared with the western medicine groups(P>0.05).2.In the study on the effect of Rhizoma drynariae and its main components in endometrium of mice model with dysfunctional embryo implantation,the expression of each index model group was lower than that of the blank group(P<0.05).In the Rhizoma drynaria and progynova groups,pinopodes were abundant.In the total flavone group and aspirin group pinopodes were low,while naringin group few.Compared with the model group,Compared with the model group,Rhizoma drynariae group had higher levels of ER,PR,LIF,and integrin ?v?3(all P<0.05).Total flavone had a similar efficacy as the whole preparation,while naringin had significant difference with Rhizoma drynariae group group for each index(P<0.05).3.In the study on the effect of Erbuzhuyu improving prescription in endometrial receptivity of patients with repeated IVF-ET failure,HE staining of the non treatment group and treatment group was not significantly different(P>0.05),but the proportion of ideal form in treatment group was higher than that in non treatment group;There was no obviously difference between the two groups of pinopodes under scanning electron microscopy(P>0.05),but the rich expression of pinopodes was higher than non treatment group;There was the obvious difference between the two groups in the expression of HOXA10mRNA(P<0.05).There were no significant differences of SCF mRNA,MMP-9 mRNA between two groups,but by the analysis of the comparison of the mean and discretization,medication group was better than non treatment group;There were the obvious differences between the two groups in the expression of LIF and integrin?v?3(P<0.05)Conclusion:1.Erbuzhuyu improving prescription might increase the expression of pinopodes,SCF mRNA,HOXA10 mRNA,ER,PR,LIF,and integrin ?v?3,conducive to blastocysts adhesion,and finally improves the endometrial receptivity.2.Rhizoma drynariae could improve endometrial receptivity of mouse models of dysfunctional embryo implantation as shown by increased numbers of pinopodes and higher levers of SCF mRNA,HOXA10 mRNA,ER,PR,LIF,and integrin ?v?3.Total flavone had a similar efficacy as the whole preparation,while naringin had a lower efficacy for all markers probably because of the dose problem.Its individual main components had variable efficacy compared with whole Rhizoma drynariae.3.Erbuzhuyu improving prescription might improve the expression of HOXA mRNA,LIF,integrin ?v?3 with mid luteal phase endometrium of patients with repeated IVF-ET failure,while it has a certain effect on pinopodes,SCFmRNA,MMP-9 mRNA.It should be affirmed that Erbuzhuyu improving prescription can improve endometrial receptivity in patients of repeated IVF-ET failures by endometrial biopsy.
