| Objective To investigate the effect of traditional chinese medicine YiKang decoctionon the adhesion of mouse endometrial cells and expression of leukemia inhibitor(LIF) and integrin-ανβ3and evaluate the inhibition efficiency of small interferingRNAs(siRNA) of LIF, and the influence on expression of integrin-ανβ3.Method All mice of4-5weeks were divided into two groups in random, witch wereintragastric administration with0.8ml YiKang decoction and0.8ml saline respectivelyfor28±7days, then serum was extracted. Mouse endometrial cells were incubatedwith Fetal bovine serum, mouse serum containing saline and YiKang decoctionrespectively. Adhesion assay was uesd to test adhesion ability of endometrial cells.The protein level of LIF in culture mediums was meaured by ELISA. The expressionof integrin ανβ3protein was analyzed by means of western blot. Meanwhile threesiRNAs sequences specifically targeting mouse LIF gene were chemicallysynthesized(named as LIFsiRNA-1ã€LIFsiRNA-2and LIFsiRNA-3).The siRNAswere transfected into primary mouse endometrial cells (MECs) by liposome. Afterselected the best clips and optimal transfection concentration, The expression ofLIF-mRNA was analyzed by means of real-time RT-PCR, The protein level of LIFin cell culture medium was meaured by ELISA, and the protein level of integrin-ανβ3was detected by Western blot. Analyzed the correlation relationship between LIF andintegrin-ανβ3on the level of protein.Results Cell adhesion ability of Medicated serum group was obviously higher thanthe control group(P<0.05). The level of LIF and integrin ανβ3protein of medicatedserum group was also significantly higher than the control group(P<0.05).WhileMECs transfected with LIF siRNA-1had significantly decreased of LIF mRNA thanthe control group (p<0.05) and decreased depending on dosage within a certain range,so70nmol/ml was selected as the optimal transfection concentration to continue theexperiment. The expression of integrin-ανβ3mRNA was significantly decreased thanthe control group (p<0.01); The level of LIF and integrin-ανβ3protein wassignificantly lower than the control group (p<0.01). LIF and integrin-ανβ3has significant positive correlation by the correlation analysis (r=0.883, p<0.01).Conclusions Yikang decoction through up-regulation of LIF promotes the expressionof integrin-ανβ3, then enhances cell adhesion function, which may play an importantrole in the process of improving uterine receptivity. |
