Part ?Tissue Factor correlation studies in invasion and metastasis of Larynx carcinomaObjective To understand the relationship between the tissue factor (TF) and laryngeal carcinoma. The difference of TF expression in pericarcinomatous tissues and carcinomatous tissues for patients with human laryngeal carcinoma was studied and its potential significance to clinic was discussed.Methods GEO data analysis, immunohistochemistry, western-blotting and RT-PCR were performed to assess the expression of TF at protein and mRNA level and the difference between pericarcinomatous tissues and carcinomatous tissues in patients (n=20) with laryngeal carcinoma was compared and analyzed.Results The results were shown that expression of TF in pericarcinomatous tissues was significantly higher than that in carcinomatous tissues (P<0.01); furthermore, the intensity of TF mRNA expression in pericarcinomatous tissues was also significantly stronger than that in carcinomatous tissue (P<0.01). Analysis of GEO dataset suggest that the expression of TF in tumor tissues has no significant difference with normal tissue (P>0.05), while lower than that of the pericarcinomatous tissues (P<0.01). In addtion, HNC patients that have lower expression of TF have longer DFS than those with higher expression of TF.Conclusion TF may contribute to the carcinogenesis and development of laryngeal carcinoma, which is an important value to judge the malignant degree and progress of laryngeal carcinoma; and it maybe a new potential target of therapeutics for human head and neck cancer.Part ?Tissue Factor correlation studies in invasion and metastasis of Larynx carcinomaObjective:To investigate the significance of exosome in invasion of HNC, understanding the mechanism of metastasis of laryngeal carcinoma.Methods We use human laryngeal carcinoma cell line Hep-2 and human endothelial cells HUVEC in vitro, adding the exosome of laryngeal carcinoma into the HUVEC, we assess the expression of TF at protein and mRNA level using immunohisto-chemistry, western-blotting and RT-PCR, moreover, we test the proliferation and migration of HUVEC using CCK-8,scarification test and transwell test?Result The results show that the exosome secreted from Hep-2, can significantly stimulate TF expression of HUVEC (P<0.01), and greatly stimulate the proliferation and migration of HUVEC (P<0.05).Conclusion TF may contribute to the carcinogenesis and development of laryngeal carcinoma, which is an important value to judge the malignant degree and progress of laryngeal carcinoma; and it maybe a new potential target of therapeutics for human head and neck cancer.
