1.Research on genetic regularity and physiological metabolism of cembratrien-diol in tobaccoCBT-diols are the main component of tobacco leaf surface exudates.The CBT-diols conten of 8306 and yuyan NO.11 were significantly higher than other varieties.The results of backcross genetic design show that the changesof CBT-diol content in fture generations are prortional to the proportion of their genetic composition in 8306.It is speculated that CBT-diols are controlled genetically,and in accordance with quantitative traits inheritance.The expression of DXR,CYC-1 and CYP71D16 genes in 8306 and yuyan NO.11 were significantly higher than other varieties.The up-regulated of DXR,CYC-1 and CYP71D16 promote the synthesis of CBT-diols.The results of cloning and sequencing show that the sequencing of CYC-1 and CYP71D16 in 8306 and 12451 are 100 % coincide with the sequencing provided by Genebank of NCBI.The amount of degradation of CBT-diols in8306 and yuyan NO.11 are higher than yunyan87 and 12451.After baking,CBT-diols degrade to solanone and derivatives,which prove an important contribution to the aroma quality of tobacco leaves.Yuyan NO.11 obtained higher sensory quality scores and industrial evaluation.2.Effect of ecological factors on the Synthesis of CBT-diols in tobaccoEcological factors have a significant effect on the Synthesis of CBT-diols in tobacco.The CBT-diols content of tobacco in Lushi was obviously higher than that in Yuzhou.In order to improve the leaf trichome exudates Yuyan NO.11 should be planted in Lushi area.This study discussed the differences of CBT-diols synthesis and expression level of CYC-1 and CYP71D16 in different temperature range and light intensity.When the temperature range was 15 ℃,the CBT-diols content and the expression level of CYC-1 and CYP71D16 were significantly higher than 0 ℃.The difference increased gradually with the passage of time.It suggests that larger temperature range is conducive to the synthesis of CBT-diols,larger temperature range lead to up-regulated of CYC-1 and CYP71D16.Higher light intensity lead to higher CBT-diols content and higher expression level of CYC-1 and CYP71D16.Tobacco plants should be cultivated in the areas where there are high temperature range and light intensity for high CBT-diols content.3.Effects of nitrogen fertilizer on trichome exudates,appearance and economic traits of high aroma flue-cured Tobacco VarietiesThe incresce of nitrogen inhibited the synthesis and accumulation of trichome secretion in yunyan NO.11.High aronma tobacco plants should be cultivated in the areas where there is poverty of the soil.Appropriate amount of nitrogen is conducive to promoting the coordination of tobacco aroma components.Varieties are the determinants of tobacco aroma quality.4.Research on physiological characteristics and curing characteristics of high aroma tobacco varietiesVarieties play a decisive role in tobacco aroma quality.It’s always the direction of the study of tobacco breeding in the world to improve the aroma quality of tobacco leaf effectively.8306 is a high aroma variety that bred starting from the tobacco trichome exudates.Compared with the conventional varieties,8306 have wider leaves in the growth period,and have higher trichome exudates,cembratrien-diol content and Petroleum ether extract content.Further more,in 8306,the neutral aroma quantity and important content of aroma substances such as Solanone were higher than that of conventional varieties.However,the density of glandular trichome in 8306 is no difference with the conventional varieties.In order to improve the baking quality of yuyan NO.11,tobacco plant individual development should be controlled.The mature standards should be appropriate to grasp when tobacco harvest.Take a high temperature during the yellowing stage.5.Improvement of the method for the extraction of leaf trichome exudatesFor improved shortcomings of traditional extraction method,a closed and circular device for the extraction of leaf trichome exudates was designed.
