[Objective] Detecting the expression of immune factors related to frequency and the expression level in peripheral blood in patients with lung cancer, and explore its correlation with the expression in lung cancer between clinical characteristics and chemotherapy response with immune factors. To provide theoretical basis for futher clinical chemotherapy combined with immune therapy.[Methods] The first part:Analysis patients with Middle-late stage lung cancer who get therapeutic with anti-tumor chemical drugs in the people’s liberation army general hospital during December2011to December2012, selected cases data and complete follow-up data before. Analysis relationship between selected patients with clinical data features and tumor stage and the effect of chemotherapy and chemotherapy side effects related to the distribution and the proportion。The second part:Tested the anti-NY-ESO-1antibody in peripheral blood serum samples from selected patients with lung cancer by enzyme-linked immunoassay detection, studies on relationship between NY-ESO-1antibody with the clinical characteristics. The third part:Textde the expression of a variety of cytokines in peripheral blood serum samples from patients with lung cancer was selected by using flow cytometry instrument of magnetic beads method and the relationship between its expression and between clinical characteristics of patients. The fourth part:To study the role of PIAS famlLy in DNA repair.To cultivate the cell line can detect the homologous and heterologous repair, to detect DNA damage repair pathway and mechanism of the gene in the PIAS famlLy, and the verify in resistance to ionizing radiation.[Results] The rate with NY-ESO-1antibody in peripheral blood was9.4%.NY ESO-1antibody in peripheral blood wad lower in patients with better chemotherapy effect,NY-ESO-1antibody level has certain correlation with clinical staging.Test results of cytokines in peripheral blood showed Increased concentrations levels of IL-17a was decreased with increased M stages.TNF-a associated with clinical effect; Only IL-4,IL-17a and IL-10; IL-6,IL-2and TNF-a were not relevant. Experiment about research the role of PIAS famILy in the DNA repair in cell lines, successful built the cell lines which can detect the homologous and heterologous repair pathway, DNA damage repair of PIAS family is carried out through two homologous and heterologous repair pathways, no synergy effect between the members of the famILy, PIAS can enhance resistance to ionizing radiation.[conclusion] NY-ESO-1antibody positive rate was higher in patients With lymph node metastasis(P=0.025). Speculated that NY-ESO-1antibodies might somehow involved lymph node metastasis. NY-ESO-1antibodies has a rising trend with recent curative effect.Although this study did not obtain statistically significant results, but the trend is worth attention. NY-ESO-1antibody and survival analysis of PFS was not are obtained statistically significant results,concentration of TNF-a of TH1type of factors associated with chemotherapy curative effect, poor effect of chemotherapy in patients with higher expression,(P=0.01). expression levels of TH1class factor increased in patients with poor effects of chemotherapy,but did not get statistically significant results, needs further study. The results indicate the better immune function in lung cancer patients are likely to achieve better results. IL-17a expression reduced in patients with distant metastasis, the results have statistical significance,(P=0.046). But this kind of change and the causal relationship between tumor metastasis remains to be further research.
