BackgroundHuman beings are susceptible to hepatitis B virus (HBV). At present, the number of infectious persons with Hepatiti B is more than300million throughout the world, and especially in China, HBV infection is even more serious. About15%-25%patients with HBV are at risk of developing into liver cirrhosis and cancer, which threaten seriously people’s health. So far, there have been no effective means to eradicate the hepatitis B. As a result, it has become a huge disaster for people’s economy and health.Aims1. To explore the level changes of AST, ALT, TBIL, DBIL, ALP, CHE in the serum of HBV makers with two pairs of semi-hepatitis B.2. To explore HBV-DNA quantitative changes in different modes of HBV serological markers.3. To explore the characteristics of frequency distribution of ApoAI-75Mspl, ApoB Msp1, ApoCⅢ Sst1, LRP5, and ApoE gene polymorphisms in different modes of HBV serological markers..Methods1. The blood samples of patients with HBV from Chuiyangliu hospital were collected randomly. Cases were diagnosed based on the required standards of the "Viral Hepatitis Prevention and Control Programme". For convenience, HBsAg, anti-HBs, HBeAg, anti-HBe, and anti-HBc were named as1,2,3,4,5respectively. The subjects were divided into9groups,1,2,3,4,5negative group;2and5positive group;1,3and5positive group;1,4, and5positive group;4and5positive group;1and5positive group;5positive group;2positive group;2,4, and5positive group. There were a total of720cases,80patients in each group,342cases were male and378female, and the average age was (42±5) years. All subjects were from the Han population in northern China without any consanguinity.2. According to the reports, the amplification primers of ApoAI-75Mspl, ApoBMsp1, ApoCⅢSst1, LRP5and ApoE genes were designed and genomic DNA were extracted.3. Detection for gene polymorphisms of ApoAI-75Msp1, ApoBMsp1, ApoCⅢSst1, LRP5and ApoE was carried out by SnaPshot method (single-base multilocus micro-sequencing), and different genotypes of each locus were conducted by the method of sequencing in order to support the final evidence of the accuracy of test results.4. Quantitative HBV-DNA was performed by fluorescent quantitative PCR method.5. Two pairs of semi-hepatitis B for HBV makers were conducted by immune uminescence.6. All the data collected from the study were analyzed by SPSS17.0and variance analysis.Results1. In the1,3, and5positive group, the level of ALT, AST, TBIL, DBIL, ALP, and CHE in the serum was obviously higher than that of the1,23,4, and5negative group(P<0.05).2. The serum level of HBV-DNA in1,3, and5positive group was higher than that of the other eight groups (P<0.05).3. There was association between gene polymorphisms of ApoAI-75Mspl and ApoE and different modes of two pairs of semi-hepatitis B (P<0.05), while there wasn’t any association between gene polymorphisms of ApoB Mspl, ApoCIII Sstl, LRP5and different modes of two pairs of semi-hepatitis B (P>0.05).ConclusionThe gene polymorphism of ApoAI-75Mspl and ApoE was associated with the different modes of HBV markers.
