Objectives:Construction-heat Syndrome IBS rat model to study the hot and humid diarrhea-type IBS rat model of colonic MC,5-HT, AQP4, PKA and cAMP changes of Jiuxiang Zhixie tablets of damp heat diarrhea type IBS mechanism.Methods:60 SD rats were randomly divided into 6 groups: blank control group, model groupⅠ(Spleen and Stomach Damp-heat group), model groupⅡ(composite model group), groupⅠ,Ⅱexperimental group, loperamide hydrochloride group, each Section 10. In addition to the blank group, the spleen and stomach in each group are to be hot and humid modeling methods, one of the model group II, Experiment II group, loperamide hydrochloride at the same time the control group was to be irritable bowel syndrome, chronic stress model method. After the success model, the rats were given different drugs or distilled water orally for 7 days, were sacrificed blood and colon tissue observation rat colon MC,5-HT, AQP4, PKA and cAMP expression of Change.Results:1. the model and experimental rats Ileocecum and distal colon of conventional HE staining no significant change; 2. ModelⅠ,ⅡIleocecum rats and distal colon 5-HT and increased MC expression, and AQP4, cAMP and PKA reduced expression, and the blank group and other treatment group difference was statistically significant (P<0.05); 3. Jiuxiang Zhixie film can improve the model rats diarrhea status, change in rat intestinal tissue in MC,5-HT, AQP4, PKA and cAMP abnormal expression, compared with blank control group no significant difference (P> 0.05); with the control drug effect considerable (P> 0.05).Conclusions:1. The success of the creation of hot and humid diarrhea type IBS rat model.2. Humid diarrhea type IBS rat model of intestinal (ileocecal, colon distal) in AQP4,5-HT and abnormal distribution of mast cells.3. cAMP-PKA signaling pathway can control AQP4 in rat intestinal expression, the impact of water within the colon of mesocaval reabsorption.4. Jiuxiang Zhixie film can improve the damp-heat diarrhea-type IBS rat model of intestinal (ileocecal, colon distal) in AQP4,5-HT and abnormal distribution of mast cells.5. Jiuxiang Zhixie film probably by influencing cAMP-PKA signaling pathway to regulate AQP4 in rat intestinal expression, the promotion of water within the colon of mesocaval reabsorption to antidiarrhea purposes.6. Jiuxiang Zhixie tablets to Jianpi Qingre dampness, Shibuya intestinal Zhixie, the analgesic gas legislation, are the treatment of damp-heat diarrhea-type IBS and the Spleen and Stomach Damp-heat diarrhea an effective prescription.
