Objective:1.To establish the quality control system of Zhizi and Zhizibopi Soup.2.To explore the mechanisam of protecting liver and improving cholereti of Zhizibopi Soup.Methods:1.To simultaneously dterminate the contents of jasminoidin and crocin-1 in gardenia using HPLC method.2. To establish the chemical fingerprints of gardenia using HPLC and HPCE method. To study the chemical pattern recognition and cluster analysis based on the fingerprints data using multivariate data analysis system.3. To analyze the genetic fingerprints of Zhizi based on inter-simple sequence repeat (ISSR) molecular marker technique and study the cluster analysis by UPGMA method to gain dendrogram.4. To study the effects of Zhizibopi Soup and it’s prescription on ALT/AST activity in serum, MDA/SOD activity in liver, injured degree of liver, bile flow and bile component, the models of CCl4 induced acute liver injury in mice were used.5. To analyze the profile-effect between fingerprints and liver protection by the method of HPLC.Results:1. The chemical fingerprint system was stability, and the peaks are well separated. It showed good similarity between the simulative mean chromatogram and sample chromatogram.2. The samples were divided in 4 clusters based on inter-simple sequence repeat (ISSR) molecular marker technique, and the samples from the same habitats could be divided in the same cluster.3. Zhizibopi Soup could reduce ALT/AST level in serum and MDA level in liver, and increase SOD activity in liver. In addition, Zhizibopi Soup could significantly increase bile flow and bilirubin content of bile, but there was no significant effect of Zhizibopi Soup on cholesterol content.4. The Zhizibopi Soup has 19 common peaks at 238nm wavelength, and 6 common peaks at 440nm wavelength. There were 4 common peaks may directly influence the functions of Zhizibopi Soup, which was marked in the fingerprints.Conclusions:1. The HPLC and HPCE fingerprints could be used to control the quality of Zhizi.2. The chemical recognition pattern based on rapid cluster analysis and Euclidean distance system cluster analysis could reflect the discrimination of Zhizi more accurary.3. Zhizibopi Soup is effective in hepatic-protective for mice and choleretic for rat. The protect liver effect are mainly ascribed to the synergic effect of Zhizi and Huangbo. The choleretic effect is mainly ascribed to Zhizi.
