ObjectiveThrough the observation influence of acupuncture in Sishencong ", " Neiguan "and" Sanyinjiao "acupoint on chronic unpredictable mild stress rats model of depression (cums) on rats weight, diet and behavior science, and through the observation of hippocampal Glu and GABA content changes, peripheral blood nerve transmitter (DA and 5-HT) levels, glutamate decarboxylase (GAD65, GAD67), glutamate transporter EAAT3, GABA transporter 1 (GAT1), NMDA receptor subunit NR2B, GABAA receptor protein expression changes of the experimental rats to explore the antidepressant mechanism of acupuncture intervention.Methods50 SPF SD rats, half male and half female, weighing 180-220g, placed 7:00-19:00 illumination,19:00-7:00 the dark laboratory, the tester checked random number table, dividing the rats into blank control group, model group, fluoxetine group, acupuncture group and non acupoint acupuncture group,10 rats in each group. Keeping the control group only 5 rats per cage, and the remaining group 4 rats per cage 1. Feeding for one week, the chronic unpredictable mild stress model of depression (cums) (measures of stress including foot shock, swimming in ice water, shaking, clamping tail, bound, water deprivation, fasting and reversed circadian, daily randomly assigned a stress average each stimulus use 2 times. Each time the needle for 30 min, 1 times a day)was applied, and in 1 days to 21 days, except the blank control group, other groups of animals was suffered from different stress, and acupuncture group was acupunctured "Sishencong", "Neiguan" and "Sanyinjiao"’ points, each time the needle for 30 minutes, once a day. Non acupoint group was acupunctured in the position near the three points of acupuncture (according to the domestic and foreign general non acupoint taking acupuncture point method, the similar two meridian line midpoint was defined as non acupoint), needle, acupuncture method, needle retaining time and period of treatment were the same as the acupuncture group. Fluoxetine group rats were given fluoxetine 1.8mg/kg weight, soluble stomach by 2M1 pure water,1 times /day. Animals in each group were weighed on into the group in the first day and the 7th day,14th day and 21st day, and amount of food intake was recorded and open field experiment was used. At the end of the experiment the tester detected hippocampal Glu and GABA content, content of peripheral blood neurotransmitter (DA and 5-HT), glutamic acid decarboxylase (GAD65, GAD67), glutamate transporter EAAT3, GABA transporter 1 (GAT1), NMDA receptor subunit NR2B, GABAA receptor protein expression.Conelus ion1. With the extension of the modeling time, body weight of rats in control group increased significantly, the treatment group significantly increased weight for fluoxetine group and acupuncture; acupuncture group increased larger than the fluoxetine group in late treatment, but no significant difference between each other (P>0.05); non acupoint group body weight increased, but the increase is slow also, increased significantly more than other rats in each treatment group, and no significant difference between model group (P>0.05); compared with the control group, the treatment group gradually close to normal levels of body weight in treatment group after 21 days of weight, but there are still some differences (P<0.05), compared with the model group and non acupoint group, difference obviously (P<0.05), each group of sucrose consumption and feed consumption changes were positively correlated with body weight. In behavioral science regulation:animals in the model group compared with normal group, the number of horizontal movement and vertical movement times were significantly reduced, significant difference (P< 0.05); acupuncture treatment group and fluoxetine group group of horizontal movement and vertical movement score compared with model group, there were significant (P< 0.05 ~0.01), and compared with the normal group, significant difference (P> 0.05) non acupoint acupuncture group and the model group had no significant difference (P> 0.05); the scores between treatment groups compared with no significant difference (P> 0.05).2. the cums rat model of hippocampal Glu content decreased significantly (P < 0.05), and GABA content increased significantly (P< 0.05), after acupuncture intervention, chronic stress depression rat model hippocampal Glu content increased significantly (P< 0.01), the GABA content decreased significantly (P< 0.01).3, CUMS rat model of peripheral blood DA content than the normal group significantly decreased (P<0.05), fluoxetine group, acupuncture group compared with the model group, the difference was statistically significant (P< 0.01). Compared with the model group, the difference was statistically significant (P< 0.05), the difference was statistically significant (P< 0.01) between the control group and the model group, the difference was statistically significant (5-HT<). Model 4, the cums rat GAD65 protein expression compared with normal control group significantly increased (P< 0.01), EAAT3 protein expression level was significantly decreased (P< 0.05). The GAT1 protein expression levels were significantly increased (P< 0.05) and NR2B protein expression level was significantly decreased (P< 0.01); and the model group, fluoxetine and acupuncture group of GAD65 protein expression level was significantly decreased (P< 0.05), EAAT3 protein expression levels were significantly increased (P< 0.01, P< 0.05). The GAT1 protein expression level was significantly decreased (P< 0.01, P< 0.05) and NR2B protein expression levels were significantly increased (P< 0.05), and non acupuncture point group had no significant difference (P> 0.05).Results1. through the intervention of acupuncture, the sucrose consumption, feed consumption and body weight were increased, and the increase of exercise activity increased, the acupuncture can relieve depression of physical and mental disorders.2. The anti depression mechanism of acupuncture is related to the regulation of dopamine (DA),5-HT content and the expression of excitatory amino acid glutamic acid and its receptor.3. The antidepressant effect of 3, with the specificity of acupoint acupuncture intervention.
