Objective:Through the observation of "Baihui" through "Qubin" effect of acupuncture on rat brain tissue CD36, HMGB1 and PKA/CREB expression in cerebral hemorrhage, to explore the brain neuroprotective mechanism.Method:a total of 150 healthy male Wistar rats, were randomly divided into three groups:model group, acupuncture group (model+acupuncture group), sham operation group,50 rats in each group. Each group was further divided into 6h,12h,24h,72h,7d five groups, to make cerebral hemorrhage model. The rats in the sham operation group underwent the operation similar to the model group, but no blood injection. The acupuncture group rats after intracerebral hemorrhage, with "Baihui" through "Qubin" acupuncture treatment. Comprehensive evaluation of rats neural function defect symptoms using Berderson score and Rosenberg score method; the expression of brain hemorrhage in a rat model at different times of the CD36, HMGB1 and PKA/CREB for the detection of brain by immunohistochemistry and Western blot methods.Result:1.Neurobehavioral:Rats in the acupuncture group compared with the model group rat nerve defect were recovered obviously, in 12h,24h,72h,7d score of acupuncture group rats were significantly lower than those in the model group, the difference was statistically significant (P<0.05).2.CD36 immunohistochemical results:In the sham group shows a small amount of brain tissue CD36 protein expression at each time point; the model group showed high expression of CD36protein at each time point, and were higher than sham group (P<0.05),acupuncture group compared with model group was significantly higher (P<0.05).3.HMGB1 immunohistochemical results:Sham group have HMGB1 weak expression; in the model group, the expression of HMGB1 protein was significantly increased, and reached the highest at 24h、72h after operation; the acupuncture group, the expression of HMGB1 protein in brain tissue at 24h,72h,7d time points were significantly increased in comparison with the model group, the difference was statistically significant (P<0.05).4.P-PKA Western blot results:each group at different time points in rat brain were P-PKA protein expression. Sham group have P-PKAweak expression; in the model group after 6h, the expression of PI3K protein was significantly increased, and reached the highest at 23h、72h after operation; the acupuncture group, the expression of P-PKA protein in brain tissue at different time points were significantly increased in comparison with the model group, the difference was statistically significant (P<0.05).5.P-CREB Western blot results:each group at different time points in rat brain were P-CREB protein expression. Sham group have P-CREB weak expression; in the model group after 6h, the expression of PI3K protein was significantly increased, and reached the highest at 23h、72h after operation; the acupuncture group, the expression of P-CREB protein in brain tissue at different time points were significantly increased in comparison with the model group, the difference was statistically significant (P<0.05).Conclusion:Acupuncture "Baihui" through "Qubin"can obviously improve the degree of cerebral hemorrhage rat model of nerve function defect score, alleviate the symptoms of nerve function defect.Acupuncture "Baihui" through "Qubin" can inhibited the expression level of CD36, HMGB1, to reduce inflammation factors on secondary injury after intracerebral hemorrhage, which may be one of the neuroprotective role mechanisms.1.Acupuncture "Baihui" through "Qubin" can increase the expression level of P-PKA, P-PKA, so as to protect the nerve cells of cerebral hemorrhage, reduce the secondary injury after intracerebral hemorrhage in rats, which may be the other the neuroprotective role mechanisms.
