ChaiHu is a commonly used traditional Chinese medicine, which was first recorded in the Classic of the Materia Medica (Shen Nong Ben Cao Jing), as a top grade. The root of Bupleunum chinense DC. and B. scorzonerifolium Willd. are the major and legal sources of Chaihu recorded in the Pharmacopoeia of People’s Republic of China. As a key player of many famous prescriptions, such as Xiao Chaihu Tang, Buzhongyiqi Tang and Xiaoyao Powder, etc., the demand of commodity increased year by year, up to3-5million kg.At present, dozens of species of Bupleurum L. used as chinese medicine "Chaihu" in different regions in China. It is contained three kinds of herb of B. marginatum Wall.ex DC, B. microcephalum Diels. and B. malconense Shan et Y. Li in the Standard for the traditional Chinese medicine in Sichuan Province, which has the same efficacy and dosage compared with the Pharmacopoeia.There differences, not only the species, but also the medicinal parts, between the local standard and the pharmacopoeia caused confusion of drug safety,effectiveness and scientificity. Because of the survey research that B. malconense is the main species of wild Chaihu products, and has adequate resources and good quality, we yielded the following results and innovation in studying the medicinal value of B.malconense.1. To study in a pharmacognostical study on the B.malconense:①The research of medical condition show that overground and underground part of different species of genus Bupleurum are medicinally in past and present.②The identification of original was studied.③Plant morphology was studied on different medicinal parts of B.malconense, to find out their main characters in macroscopic and microscopic identification.④Identification of TLC, physical and chemical were studied, and then the limit standard of content of moisture, ash and extract were established.⑤As the main active ingredient, saikosaponin a, c, d were determined by HPLC. We found that the content of saikosaponin in the underground part can meet the requirements of Pharmacopoeia, but the content of the aerial part is very low, only1/10of the former. 2. The paper first reported comparative study of pharmacodynamics and acute toxicity test of different medicinal parts of B.malconense. Because of reducing the level of GOT, GTP and MDA, increasing the level of GSH, reducing the edema and degeneration of liver cell, protecting the hepatic injury caused by CCl4in mice and reducing fever induced by yeast in rats, the two part of B.malconense have some antipyretic and hepatoprotective effect. The water extract in high dose has a certain degree of acute liver toxicity, which can return to normal after1weeks stoping drug3. The volatile constituents of B.malconense and B. Chinense. were analyzed, using AMDIS and retention index for the fist time. We were identified47,45and50kinds of volatile components from the underground, overground part of B.malconense and root of B. chinense. Results show that B.malconense have a certain scientific basis for treating cold fever disease, as the main components have good consistency, although the different species and relative content of volatile constituents.4. For the first time, the isolation and structure identification of the chemical constituents from the aerial part of B.malconense.22constituents were isolated from the ground part of ethyl acetate extraction, and then12compounds were identified by spectroscopic techniques and the physical and chemical properties which were composed of flavonoids and so on. All of them were isolated from the plant of B.malconense. for the first time, and3compounds from Bupleurum L. for the first time, which has1new compound.5. Firstly, the fingerprints of flavonoids of overground part of B.malconense. were established by HPLC, which identified9common peaks, choose peak4(rutin) as reference peak (S), calibrated6peaks, and stipulated the similarity and relative retention time of the characteristic peaks. Then, methods of the content of total flavonoids were established by HPLC for the fist time, that determination of the three flavone glycoside. The results of the study show that the content of total flavonoids overground part of B.malconense. is about4%, furthermore we may safely draw the conclusion that it has very good pharmaceutical value. 6. The quality standard (Draft) of aerial part and root of B.malconense. were established respectively, to provide a reference for quality control of the herb.This thesis makes systematic study on the aerial and underground part of B.malconense from the pharmacognosy, chemical constituents, pharmacological effects, quality standard etc. We can draw the conclusion that the different parts of B.malconense should be used as medicines separately, for there were great differences in the content of pharmacodynamic material of volatile oil and total flavonoids. Pharmacodynamic substance foundation of ground part are studied, that12compounds were authenticated, on the basis of which, flavonoids fingerprint and determination of total flavonoids were established, and then quality standard (draft) of different part are formulated respectively. The above work provides a scientific basis for the respectively use of aerial part and root of B.malconense, and laid the foundation for rational utilization of the resources.
