Aim:To establish of infections caused by herpes simplex virus type I in mice animal models of Peripheral facial nerve palsy (FNP) with the compatibility of distal-proximal acupoints in acupuncture treatment. To observed the therapeutic effect of acupuncture on FNP by useing Jiache (ST6) and Hegu (LI4) Points. Intending to reveal the nuclear transcription factor-kb (nf-kb) molecular signaling mechanisms with the acupuncture treatment with the using of compatibility of distal-proximal acupointsfor treating the FNP.Providing the theoretical evidence of selecting points for the clinical treatment of FNP.Methods:We Selected120mice, male and female, weighing18.5-20.5g provided by Chengdu Suoda Bio-Technology Co.,Ltd.There were four groups with30mice in each:Group A, blank control group; Group B, saline group; Group C, model animal group; and Group D, electro-acupuncture group.Each group was further divided into Three time points (3days、7days、14days)groups. Each time point has10mice.Dropping the virus in the nerve stump with the Model group and electro-acupuncture group.Saline group added dropwise saline as a control, through Model evaluation,the presence facial paralysis in mice Handled in accordance with the corresponding grouping, electro-acupuncture group Carry on the Acupuncture and electro-acupuncture treatment,theBlank group,Saline group, model group were fixed without any treatment, electro-acupuncture group The local Selected Jiache (ST6) distal Selected Hegu (LI4). Jiache (ST6) and Hegu (LI4) were Both treated with reinforcing-reducing method. Electro-acupuncture was given by a G6805after fixing the mice.The stimulation frequency was3-4Hz and intensity was1-2V in continuous wave (CW) mode to make the needle slightly vibrate and keep the mice quiet. The needle was retained for20min, once a day, and the treatment lasted for3days,7days and14days.Ten mice in each group were anesthetized by intraperitoneal injection of sodium pentobarbital (50mg/kg) and killed by decollation quickly after3,7, and14days of electro-acupuncture. The intratemporal portions of the right facial nerves, geniculate ganglia, brainstem, and cerebral cortex were dissected. HSV-1DNA was detected in the model animal group and electro-acupuncture group. However, the quantity of HSV-1was significantly lower in the electro-acupuncture group compared with the model animal group (P<0.05after3days, P<0.01after7and14days). After7days of acupuncture, HSV-1DNA was decreasing in the electro-acupuncture group, while HSV-1DNA was still higher compared with4days prior in the model animal group. HSV-1DNA in the model animal group was decreasing14days after acupuncture.3.The expression of nuclear transcription factor-KB (NF-κB)The expression of NF-κB began to increase significantly after3days in Model group, the score gradually reaches a peak after7days,but decreased after14days which still significantly higher than the normal group and the saline group.Compared with the model group, the quantity of NF-κB was significantly lower in the electro-acupuncture group compared with the model animal group after3,7, and14days of lectro-acupuncture (P<0.05)4.The expression of IκBα proteinThe expression of IκBα Significantly decreased after3days with Virus infection in mice, the expression of IκBα is still at a low level after7days, but increased after14days which still significantly lower than the normal group and the saline group. Compared with the model group, the quantity of IκBα was significantly higher in the electro-acupuncture group compared with the model animal group after3,7, and14days of lectro-acupuncture (P<0.05)5.The expression of IL-1β and TNF-αThe expression of IL-1β and TNF-a began to increase significantly after3days in Model group(P<0.05), the score gradually reaches a peak after7days.but decreased after14days which still significantly higher than the normal group and the saline group.Compared with the model group, the quantity of IL-1β and TNF-α was significantly lower in the electro-acupuncture group compared with the model animal group after3,7, and14days of lectro-acupuncture (P<0.05) Conclusion:1.NF-κB、NF-xBmRNA、IL-1β and TNF-α is positive related with the degree of FNP in NF-κB signal pathway.2.The method using distal-proximal points can alleviate FNP symptoms. The electro-acupuncture treatment Can Make the FNP Effect indicators,Neurons,Schwann cells of blood Tending to be normal By adjust the NF-κB,NF-κBmRNA, IκBα, IL-1β, TNF-a to Reach the Purposes of Anti-inflammatory.3.The treatment of using distal-proximal points FNP Has Efficacy Of Cumulative effect, Electroacupuncture through Longer Period Stimulating meridian can make hsv-1, IL-1β, TNF-α Continued Reducting,thereby to the Purposes of Anti-inflammatory and Anti-infection More Efficacy Significantly4.About all the Conclusion above Prompting that the method using distal-proximal points is Effective In the acute phase of FNP through regulating NF-κB signaling pathway. And with the extension of Period tends to be the best therapeutic effect.
