| Objective:To observe the best ratio of hypericum attenuatum choisy and Astragalus mongholicus for the protective effects against myocardial ischemia-reperfusion injury in rats, and explore the mechanism against myocardial ischemia-reperfusion injury.Methods:1.Combinate the hypericum attenuatum choisy and Astragalus mongholicus according to 1:1,1:2,2:1 Effect on myocardial ischemia-reperfusion injury rats by these matches.Observe the CK content and range of myocardial infarction of model rats, screen the the active site against myocardial ischemia reperfusion injury.2.Observe the effect on pathological morphology and ultrastructure of myocardial injury in model rats through the light microscope, and instruct the myocardial protection in rats.Observe the effect on the content of myocardial apoptotic index of MIRI rats.3. Observe the effect on the content of SOD, MDA, the expression of caspase-3 and TNF-a, the expression of Hsp70 and NF-κBp65, and explore the mechanism against myocardial ischemia reperfusion injury.Results:1. The hypericum attenuatum choisy and Astragalus mongholicus ratio at 1:1 and 1:2 can obviously decrease the CK content and range of myocardial infarction of model rats,1:2 can improve pathological morphology and ultrastructure of myocardial injury in rats.2. The hypericum attenuatum choisy and Astragalus mongholicus ratio at 1:2 can obviously decrease the apoptotic index of MIRI rats.3. The hypericum attenuatum choisy and Astragalus mongholicus ratio at 1:2 can obviously decrease the content of MDA,1:2 dand 1:1 matching can increase the vitality of SOD.4. The hypericum attenuatum choisy and Astragalus mongholicus ratio at 1:2 can significantly cut the caspase-3 protein expression, cut TNF-a protein expression. It also can decrease the caspase-3 mRNA expression, cut TNF-amRNA expression.5. The hypericum attenuatum choisy and Astragalus mongholicus ratio at 1:2 can significantly cut the NF-κB p65 protein expression, raise HSP70protein expression. It also can decrease the NF-κB p65 mRNA expression, raise HSP70 mRNA expression.Conclusions:1. The hypericum attenuatum choisy and Astragalus mongholicus ratio at 1:1 and 1:2 can obviously decrease the CK content and decrease range of myocardial infarction of model rats, The hypericum attenuatum choisy and Astragalus mongholicus ratio at 1:2 can improve pathological morphology and ultrastructure of myocardial injury in rats, and they have the protective effects against myocardial ischemia-reperfusion injury in rats.2. The hypericum attenuatum choisy and Astragalus mongholicus ratio at 1:2 can obviously decrease the apoptotic index of MIRI rats.3.The hypericum attenuatum choisy and Astragalus mongholicus ratio at 1:2 can obviously decrease the content of MDA,1:1,1:2 matching can increase the vitality of SOD.The mechanism may be related to inhibition of lipid peroxidation and improve antioxidant capacity.4. The hypericum attenuatum choisy and Astragalus mongholicus ratio at 1:2 can significantly cut the caspase-3 protein expression, raise TNF-α protein expression. They also decrease the caspase-3 mRNA expression, cut TNF-α mRNA expression. They probably play the role of protecting myocardial ischemia-reperfusion injury by inhibiting apoptosis.5. The hypericum attenuatum choisy and Astragalus mongholicus ratio at 1:2 can significantly cut the NF-κB p65 protein expression, raise HSP70 protein expression. Also can decrease the NF-κB p65 mRNA expression, raise HSP70 mRNA expression.They protect the injury tissues by cut the singal pathway of NF-κB p65. |
