ObjectiveTo establish depression sleep disorders (SDD) rat models, then explore the effect of acupuncture, anti-depressant and acupuncture combined with medicine on SDD rats:1. weight and behavioral changes; 2. hypothalamus, amygdala, dorsal raphe 5-HIAA/5-HT ratio; 3. NRD 5-HT1A and 5-HT2A Receptor mRNA Expression; 4. βCaMKII protein Expression on LHb; 5. Hippocampus and Serum SOD Activity and MDA content changes.Method8 normal Sprague Dawley rats were caged together normally without any stimulus and disposal.32 Sprague Dawley rats were underwent a 21d solitary raised+CUMS+REM sleep deprivation method modeling.28 rats successfully completed the modeling, and divided into model group, acupuncture group, medicine group, acupuncture with medicine group, each group with 7 rats. Model group rats, only modeling without intervention. Acupuncture group rats were acupunctured on acupoint Yintang, Shenting and Taichong(both) each day for 20min with a 15s stimulation every 10 min. Medicine group rats were gavaged administration with Zoloft and Alprazolam solution. Acupuncture with medicine group combined acupuncture and medicine intervention. Three intervention groups were given intervention for 7 days.Result1. Body weight, Open-field scores and sucrose preferences of modeling rats were significantly changed (P<0.05) compared with the normal group.2. At 28d, body weight and Open-field scores of each intervention group were significantly improved compared with the model group (P<0.05), wherein Open-field scores of acupuncture with medicine group enhanced significantly compared with acupuncture group and medicine group.3. Only sucrose preference of acupuncture with medicine group improved significantly compared with both model group, acupuncture group and medicine group (P<0.05).4. SDD modeling on rat hypothalamus, amygdala, dorsal raphe 5-HIAA/5-HT ratios have shown a significant reduction (P<0.05); only the acupuncture group of hypothalamic 5-HIAA/5-HT the ratio was significantly increased (P<0.05), acupuncture group amygdala 5-HIAA/5-HT ratio was significantly increased (P<0.05), the various brain regions other intervention group 5-HIAA/5-HT ratio compared with the model group does not appear obvious changes (P>0.05).5. Only the 5-HT1AR mRN A expression of acupuncture with medicine group was improved significantly (.P<0.05); the 5-HT2AR mRNA expression of the three intervention group were reduced significantly compared with the model group (P<0.05).6. Each intervention groups’LHb βCaMKII expression were downregulated significantly compared with the model group (P<0.05), the effect of acupuncture group and medicine group is quite (P>0.05), acupuncture with medicine group was siginificantly better than acupuncture group and medicine group (P<0.05).7. SOD activity of hippocampal tissue and serum of SDD rats was significantly lower than the normal rats(P<0.05), and hippocampus tissue and serum MDA content significantly increased than the normal rats(.P<0.05); Acupuncture group’s SOD activity of hippocampal tissue and serum were upregulated than the model group(P<0.05), and only acupuncture group’s MDA content in hippocampus has been significantly downregulated (P<0.05); Medicine group’s SOD activity of serum were downregulated(P<0.05), others were not; Acupuncture combined with medicine group’s SOD activity of hippocampal tissue and serum were upregulated(P<0.05), hippocampus tissue and serum MDA content were not downregulated(P>0.05).Conclusion1. SDD modeling can significantly change the weight and behavior of rats, acupuncture, anti-depressant, acupuncture combined with medicine can significantly improvement in its role, which acupuncture combined with medicine has the best effect.2. SDD modeling impact the sleep in rats with 5-HT-related nervous system, acupuncture and acupuncture combined with medicine intervention can play a role in improving 5-HT on the level of activity.3.5-HT receptor studies suggest that:acupuncture combined with antidepressant has a fast onset, and can significantly regulate the problem of antidepressant which with slow onset and exacerbation the depression in two weeks.4. Acupuncture is effective on SDD, with a similar effect of medicine group; acupuncture with medicine group has a better effects, deserves to be further explored and studied in clinical practice.5. Sleep deprivation depression modeling can significantly accelerate aging; 3 ways of 7d interventions on SDD rats have some anti-aging effects. And acupuncture group has the best effect with acupuncture combined with medicine group followed.
