Study On The Correlation Of Single Nucleotide Polymorphism Of PDE4D And ALOX5AP Gene With Cerebral Infarction In Xinjiang Uygur And Han

Objective:To investigate the relationship of the single nucleotide polymorphisms of phosphodiesterase4D (PDE4D) gene and5-lipoxygenaseactivatingprotein (ALOX5AP) gene and the main risk factors with cerebral infarction (CI) in uygur and han patients of Xinjiang. Methods:790CI patients and790healthy controls are collected in Xinjiang. Polymorphisms of rs11956001、rs13160982、rs829259、rs10035437、rs966221、rs918592in PDE4D and rs17216473、rs4073259、rs17222919、rs9579646、rs10507391in ALOX5AP gene were determined by SNaPshot minisequencing method. Each genotype frequency and allele frequency was analyzed and compared. Results:Section one:1) Clinical data analysis found that there were significant association with body mass index (BMI), smoking history, drinking history, hypertension, hyperlipidemia and cerebral infarction (P<0.05);2) BMI and TG in Uygur patients was higher than that in Han patients (P<0.05). Hypertension and diabetes mellitus in Han patients was higher than that in Uygur patients ((P<0.05).3) TOAST of CI showed that LAA (37.5%) in Uygur group and SAA (47.6%) in Han group (P<0.05). Section two:1) There was statistical difference in the distribution of genotypes and allele frequency of rs966221and rs829259loci of PDE4D gene between CI group and control group (P<0.05). The frequencies of rs966221genetype CC and C allele in CI group were49.9%,70.7%and46.7%,67.2%in control group (P<0.05). After adjusting age, gender, nationality, smoking history, drinking history, hypertension, hyperlipidemia, There was statistical difference in the distribution of CC genotype frequency of rs966221(CC versus TT:OR=1.551;95%CI:1.058-2.274, P=0.024). The frequencies of rs829259genetype CC and C allele in CI group were46.2%,67.4%and40.0%,67.2%in control group (P<0.05). After adjusting age, gender, nationality, smoking history, drinking history, hypertension, hyperlipidemia, There was statistical difference in the distribution of AA genotype frequency of rs829259(AA versus TT:OR=1.456;95%CI:1.001-2.119, P=0.049).2) Divided by nationality, the frequencies of CC genotype of rs966221in Han patients (65.6%) is higher than that in Han controls (62.3%), which was statistical difference (P<0.05); After adjusting age, gender, smoking history, drinking history, hypertension, hyperlipidemia, There was statistical difference in the distribution of genotype frequency of rs966221(CC versus TT: OR=3.617;95%CI:1.445-9.051P=0.006; CT versus TT:OR=3.566;95%CI:1.454-8.744P=0.005). The frequencies of AA genotype of rs918592in Uygur patients (19.0%) is higher than that in Uygur controls (12.9%), which was statistical difference (P<0.05); After adjusting age, gender, smoking history, drinking history, hypertension, hyperlipidemia, There was not statistical difference (P>0.05).3) Divided by gender, the frequencies of genotypes and alleles of the rs918592in Uyger female patients are higher than that in Uygur female controls (P<0.05), After adjusting age, nationality, smoking history, drinking history, hypertension, hyperlipidemia, There was statistical difference in the distribution of genotype AAfrequency of rs918592(AA versusGG:OR=2.433;95%CI:1.008-5.876P=0.048).4) Divided by TOAST, There was statistical differencein the distribution of genotypes frequency of rs966221between LAA and SAA group.(P>0.05) Section three:1) There was statistical difference in the distribution of genotypes and allele frequency of rs10507391of ALOX5AP gene between CI group and control group (P<0.05). After adjusting age, gender, nationality, smoking history, drinking history, hypertension, hyperlipidemia, There was statistical difference in the distribution of CC genotype frequency ofl0507391(AA versusTT:OR=1.683;95%CI:1.223-2.316; P=0.001).2) Divided by nationality, the frequencies of CC genotype of rs10507391in Han patients (22.0%) is higher than that in Han controls (14.4%), which was statistical difference (P<0.05); After adjusting age, gender, smoking history, drinking history, hypertension, hyperlipidemia, There was statistical difference in the distribution of genotype frequency of rs10507391(AA versus TT:OR=2.346;95%C1:1.507-3.651P=0.000).3) Divided by gender, the frequencies of genotypes and alleles of the rs10507391in male patients are higher than that in male controls (P<0.05), After adjusting age, nationality, smoking history, drinking history, hypertension, hyperlipidemia, There was statistical difference in the distribution of genotype AAfrequency of rs10507391(AA versusTT:OR=1.969;95%CI:1.302-2.976P=0.001).5) Divided by TOAST, There was not statistical differencein the distribution of genotypes frequency of all SNPs of ALOX5AP between LAA and SAA group.(P>0.05). Conclusion:Hypertension, hyperlipidemia, smoking, drinking history, body mass index are the major risk factors for cerebral infarction. rs966221CC and rs829259AA of PDE4D gene and rs10507391AA of ALOX5AP gene may be the susceptibility genotypies of cerebral infarction in patients of CI in Xinjiang. rs918592AA gegetype and A allele of PDE4D gene might be an association with female Uygur patients of cerebral infarction...
