The Study Of Jiangzhi Xiaoban Tablet To Cure Patients With Carotid Artery Plaque And To Influence The Plaque In Apoe Knockout Mouse As Model

Objective:To observe the clinical effects of Jiangzhi xiaoban tablet on patients with carotid artery plaque and to explore the influence of Jiangzhi xiaoban tablet on ApoE knockout mouse AS models and its related mechanisms.Method:1.Patients consistent with the inclusion criteria were randomly divided into the treated group which was treated with Jiangzhi xiaoban tablet and the control group treated with atorvastatincalcuim tablet,and then after3months the curative effects of syndromes of TCM, level of blood lipids and CRP, IMT of carotid and integration results carotid artery plaque were observed.2.ApoE knockout mouse AS models were randomly divided into treated group which was treated with Jiangzhi xiaoban tablet and control group treated with simvastatin tablet,and normal control group was set with C57BL/6J mice,all by intragastric administration,and then after12weeks plaque pathology,level of lipids,hs-CRP,TNF-a,NO,MDA,and immunohistochemical detection of CD68,a-SMA,MMP-9,NF-κB were observed.Result:Jiangzhi xiaoban tablet could significantly reduce scores of TCM syndromes with the total effective rate of96.67%for patients with carotid artery plaque,and in these areas of improving syndrome of TCM,reducing the levels of TC、TG、LDL-C、CRP,increasing level of HDL-C,and reducing IMT of carotid and plaque score,The therapeutic effects of Jiangzhi xiaoban tablet were significantly superior to control group(P<0.05).And Jiangzhi xiaoban tablet could effectively intervent with carotid plaque of ApoE knockout mouse AS models by pathological observation,and the area ratio was reduced compared with the model group(P<0.01),besides,the levels of TC、TG、LDL-C、hs-CRP、TNF-a and MDA were reduced,and the level of NO was increased(P<0.05or P<0.01),Most noteworthy is the treated group was superior to the control group with simvastatin in the way of reducing the levels of hs-CRP and TNF-a(P<0.05).And Jiangzhi xiaoban tablet could stabilize carotid plaque by reducing expression of CD68,MMP-9and NF-KB,and increasing expression of a-SMA(P<0.05or P<0.01),and superior to the control group with simvastatin in the way of reducing the expression of NF-κB(P<0.05).Conclusion: Jiangzhi xiaoban tablet not only can relieve the clinical symptoms for patients with carotid artery plaque,but also regulate the blood-lipid and reduce the levels of inflammatary factor CRP,IMT of carotid and plaque score.And Jiangzhi xiaoban tablet can regulate lipid metabolism by reducing the levels of TC、TG、 LDL-C,suppress inflammatory response by reducing the levels of hs-CRP,TNF-a and NF-KB,improve the brachial artery endothelial function with high level of NO,have anti-oxidative effect with low level of MDA,and stabilize and eliminate carotid plaque by reducing expression of CD68, MMP-9and NF-κB and increasing expression of a-SMA for ApoE knockout mouse AS models.
