Objective:To study on the therapeutic effect and the mechanism of the FFJQC capsule on CP and BPH.Method:(1) experimental animal model of prostatitis:respectively inject25%Xiaozhiling Injection into male rat prostate dorsal lobe and1%carrageenan into the male rat prostate ventral lobe to induce non-bacterial CP rats model.3.0×108/ml of Staphylococcus aureus and Escherichia coli mixed bacteria injected into the male rat prostate ventral lobe to induce in chronic bacterial prostatitis model, then observe the morphology of prostate, calculate the prostatic index, Leukocytes and lecithin corpuscles in pretherapy, observing the pathological changes of the prostate tissue with Light, detect serum proinflammatory cytokine levels with the ELISA.(2) experimental animal model of prostatic hyperplasia:castrated male mice were injected subcutaneously with testosterone propionate-induced proliferation of prostate tissue. Visual observation of prostate morphology, the calculation of prostate index,under the light microscope to observe the pathological changes of the prostate tissue, PCNA, VEGF, FGF-2and the level of FAS expression, TUNEL staining detection by immunohistochemical staining in rat prostate tissue of rat prostate cells apoptosis rate, measured T and E2concentration in serum by radioimmunoassay.(3) other pharmacodynamics:experimental determination of the internal pressure in the rat bladder intravesical pressure changes and in the value of growth inhibition rate of bladder pressure. Micro circulation in mice experimentally determined microcirculation arterioles (A), thin vein blood flow velocity (V), vascular caliber, open capillaries. The experimental determination of writhing and calculate the analgesia inhibitory rate.Results:(1) experimental animal model of prostatitis:FFJQC capsule in the reagent volume within the spirit of extinction mole rats induced by chronic prostatitis model has a significant protective effect. It can maintain normal prostate morphology, reduce prostatic index, decrease the number of leukocytes in the prostate tissue, increase the number of lecithin in the prostate tissue, anti-chronic inflammatory cell infiltration, anti-fibrosis, and helping the recovery of the prostate secretory function. Its mechanism of action associated with the lower levels of sICAM-1synthesis and expression, reduce the secretion of MCP-1, inhibition of pro inflammatory cytokines IL-1β, IL-6, TNF-alpha, IL-2induced chronic proliferative inflammation. Rat model of chronic prostatitis caused by carrageenan has a significant protective effect. It can maintain normal prostate morphology, reduce prostate index, reduce prostate edema and inflammatory cell infiltration, reduce the gland degeneration and necrosis, aseptic inflammatory reaction lower than the model control group. Chronic prostatitis model in rats caused by bacteria has reduced the role of the degree of response of prostatitis.(2) experimental animal model of prostatic hyperplasia:FFJQC capsule in the reagent volume due to the model of mouse prostatic hyperplasia caused by testosterone propionate has a significant protective effect. It can reduce the volume of the prostate and reduce prostate index, maintain the normal shape of the prostate tissue, improve the degree of proliferation of glandular epithelium, smooth muscle hyperplasia, and the interstitial congestion, edema. Its mechanism of action associates with suppressing the expression of PCNA, VEGF,FGF-2and increasing the expression of FAS,apoptosis rates in the prostate tissue,but has nothing to do with the T, E2levels in serum.(3)other pharmaco dynamics:FFJQC capsule in the scope of the reagent volume can reduce the internal pressure of the rat bladder, improve the mouse’s auricle circulation, prolonged clotting time, reduce the xylene-induced mouse ear swelling and swelling degree of inhibition rate, reduce the number of writhing response in mice and improve analgesic suppression rate.Conclusion:FFJQC capsule in the reagent volume within the spirit of extinction mole rats induced by chronic prostatitis has a significant protective effect, its mechanism of action associated with the lower level of sICAM-1synthesis and expression, reduce the secretion of MCP-1, inhibition of pro inflammatory cytokines IL-1β IL-6, TNF-alpha, IL-2induced chronic proliferative inflammation-related; It has a significant protective effect on rat model of chronic prostatitis caused by carrageenan, mixed bacteria,it can reduce prostate inflammation reaction; the rats and mice model of prostate hyperplasia caused by testosterone propionate have a significant protective effect, and its mechanism associated with the lower level of PCNA, VEGF and the expression of FGF-2in the prostate tissue, increased expression of FAS in prostate tissue and cell apoptosis rates; It can reduce the internal pressure in the rat bladder and promote urination, improve micro circulation, reduce edema and peripheral pain caused by inflammatory substances.
