Daintain/AIF-1, a novel inflammatory factor, functions as the activation ofmacrophage. It has been reported that Daintain/AIF-1could promote the proliferation andmigration of vascular smooth muscle cell (VSMC), thus, resulting in the formation ofatherosclerosis (AS). Daintain/AIF-1has also been found to play a role in many autoimmune diseases. In our previous study, Daintain/AIF-1has been confirmed to acceleratethe proliferation and migration of breast cancer cells, which first reveals the relationbetween Daintain/AIF-1and cancer and elucidates the founctions of Daintain/AIF-1onthe inflammation-cancer chain.Hepatocellular carcinoma (HCC) is a typical inflammatory correlative disease.Some researchers have reported that Daintain/AIF-1positive expressed activated kupffercells locating in the lining walls of liver sinusoids. Therefore, we hypothesized thatdaintain/AIF-1might participate in the development of HCC. In the present study,Daintain/AIF-1was found to be benefit to the formation of tumor microenvironment.Daintain/AIF-1also promotes the proliferation of HepG2cells and accelerates the immuneescape of HepG2cells. Additionaly, Daintain/AIF-1enhanced the resistance of HepG2cells to cisplatin.The major contents and results of this study as follows:1) In our study, we constructed stability RAW264.7cell lines in which overexpressDaintain/AIF-1and investigated the effect of Daintain/AIF-1on the production of NO andROS. We also examined the effect of Daintain/AIF-1on the gene expression ofchemokines.2) In the present study, Daintain/AIF-1was found to not only directly promote theproliferation and migration of HepG2cells, but also indirectly accelerate the proliferationand migration of HepG2cells via activation of IGFs/IGF-1axis.3) Daintain/AIF-1promotes the upregulation of FasL expression on the surface of HepG2cells, subsequently induces the apoptosis of co-incubated T lymphocyte,eventually, resulting in the production of immune escape.4) Combinatorial application of Daintain/AIF-1and cisplatin accelerate theresistance of HepG2cells to cisplatin.Taken together, our study showed that Daintain/AIF-1might play an important role inthe progression of HCC.
