Daintain is a protein of 146 amino acid residues isolated from porcine intestine in 1994. The next year, Daintain was identified from rat and was termed as Allograft Inflammatory Factor-1. According to their homology, we name them Daintain/AIF-1. Some experiments have proved that Daintain/AIF-1 can promote proliferation of vascular smooth muscle cells, in this way Daintain/AIF-1 is strongly correlated with cardiac allograft casculopathy.According to the previous experiments in our lab, it has been discovered that the expression of Daintain/AIF-1 levels are up-regulated and correlate with the grades of breast tumors from benign to malignant phases, suggesting that Daintain/AIF-1 is, as a risk factor, potentially involved in breast carcinoma development and progression.To study the effect of Daintain/AIF-1 on the breast cancer cell lineage MDA-MB-231, we set two testing groups. For the first group, Daintain/AIF-1 was stably transfected with breast cancer cell MDA-MB-231. After detection by Western blot and RT-PCR, transfectants were cultured and proliferation assay was performed. For the second group, MDA-MB-231 were cultured in serum-free medium containing 1μg/ml Daintain/AIF-1 and counted by standard hemocytometer.For the first testing group the results demonstrate that breast cancer cells transfected with Daintain/AIF-1 grew at a more rapid rate than the control group cells. For the second group, the proliferation results demonstrate that breast cancer cells with Daintain/AIF-1 in serum-free cell culture medium, the proliferation compared with untreated cells was enhanced.So we find out one relationship between Daintain/AIF-1 and breast cancer cells MDA-MB-231 is that Daintain/AIF-1 has the function of promoting proliferation on MDA-MB-231. This may suggest that Daintain/AIF-1 could affect the development and progression of breast cancer by enhancing the proliferation of breast cancer cells.
