Objective To study the microsatellitepolymorphism of tumor necrosis factor P gene(TNFc)In Grav es' Disease, to analyse the association between TNFc polymor phism and the susceptibility in Graves' Disease, and to further explore the mechanism in Graves' Disease. Methods 36 Graves' Disease patients and 36 healthy controls were included in the study.The TNFc microsatellite polymorphi sm which lied in the first intron of tumor necrosis factor P gene was amplified by polymerase chain reaction. We analysed all the samples' allele frequency and genotype frequency by polyacrylamide gel electrophoresis and silver staining. Results TNFc microsatel1ite polymorphism had two alleles(TNFclandTNFc2)and three kinds of genotype: homozygote TNFclcl(179/179)?TNFc2c2(181/181)and heteroz ygote TNFclc2(179/181). The TNFc2 allele frequency in Graves' Disease patients and healthy controles was respect ively 29% and 15%. Conclusions The TNFc2 allele frequency in Graves' Disease groupe was greater than cont role groupe (X2=4.02,P<0.05) which indicated an associati on between the TNFc2 allele and the susceptibity in Graves' Diseas e .No significant difference was observed in the TNFclcl genotype frequency between the Graves' Disease patients and the healthy controles (X 2=2. 72, p>0. 05) .
