China is the world’s first pig-raising country, the average quantity of amount of livestock on hand2011about478000000head,692000000head of total output, accounting for the world total pig slaughter51%, pork consumption accounted for more than60%of the total amount of China’s meat consumption, development of pig industry is directly related to the improved living standards and food safety of meat. Our current implementation rate of artificial insemination is60%, and is increasing at an average annual rate of5%, promotion and application of artificial insemination techniques has become the new century the development of pig industry in China the most biological technology key. But with the rapid development of pig industry to scale and modern production mode, integrated viral infections in reproductive and respiratory syndrome disease makes the pig industry in China increasingly complex and severe tendency, porcine reproductive disease virus infection rate has accounted for more than60%of pig disease. Continuing outbreak of reproductive disorders the modernization swine industry suffered a major setback and brings the enormous economic losses to the pig industry, severe epidemic situation the risk risk pig diseases in China has been at the forefront of the world, have posed a serious threat to the pig industry development and meat product safety in china.Pathogenic viruses can carry through semen by means of artificial insemination in the pig industry widely spread.Research on the key technology of artificial insemination for unique tropical climate characteristics of Hainan, preparation of semen dilution with physiological characteristics of sows, appropriate selection of antibacterial semen dilution with tropical semen preservation method, efficient semen diluents for the control of pig virus transmitted diseases, improve the sow reproductive conception rate, large area preventing the spread of disease, enhance the overall development ability, the pig industry to prevent large area major outbreaks of disease, improve the safety of the production and supply of meat dish, to ensure the health of the national life all has very important positive significance, no provision has an important role in construction of specified animal disease free zone and base of livestock products export countries in Hainan province.The application of the PCR method, to375boar semen from9pig farms for5types of virus have been surveyed in Hainan province. The results showed that the porcine reproductive and respiratory syndrome virus (PRRSV) positive rate was 11.47%, pseudorabies Virus (PRV) samples, the positive rate was4.80%,5related to reproductive disease virus sampling the total positive rate of samples was28.53%, Hainan in recent years, PRV and PRRSV occurrence with the increasingly complex and boar semen virus spread.By comparing the experimental optimization, propose an efficient basis for swine artificial insemination diluents formula:each1000ml double distilled water, glucose15.50g, tri-sodium citrate11.65g, EDTA2.35g, sodium bicarbonate1.75g, polyvinyl alcohol (PVP, Type11)1.00g, three carboxyl methyl amino methane (Tris)5.50g, citric acid4.10g, cysteamine acid0.07g, vitamin C1.0g. Sperm in vitro temperature preservation time than Hainan pig enterprise original common dilution to extend1.5days.Porcine semen diluents added antimicrobial effect comparison test results of different antibacterial drugs, enrofloxacin in0.1mg/ml concentration could inhibit the semen in the effect of bacteria, i.e. enrofloxacin in boar semen dilution in the inhibition effect is the best. Porcine semen diluents added antimicrobial effect comparison test results of different antibacterial agents display, semen dilution of miconazole in0.1mg/ml concentration could inhibit the semen of the effect of fungi, namely, miconazole in boar semen dilution antifungal effect. The basis of dilution in the optimization of adding antibacterial enrofloxacin and miconazole screening can make sperm in vitro effectively save time than Hainan pig enterprise with ordinary dilution to extend2day to5day sperm in vitro, effective storage period.Using7kinds of different antiviral drugs on preservation of boar semen at the same temperature, dilution conditions that effect:Yinhuang preparations and Ribavirin dilution in a concentration gradient of0.01mg/ml with the application of Yorkshire, Duroc and Landrace pigs semen preservation effect is best. Efficient semen sperm quality of preservation and Hainan pig enterprise with ordinary dilution, semen storage time increased from3day to6day.To study the effects of7different antiviral dilutions on activities of antioxidant enzymes in porcine semen. In addition to acyclovir, antioxidant capacity of Yinhuang etc.5kinds of Chinese traditional medicine and chemical medicine Ribavirin could significantly increase the sperm in the semen dilution during storage. Efficient semen form Yinhuang and Ribavirin combination diluents, has anti-pseudorabies virus (PRV) and porcine reproductive and respiratory syndrome (PRRSV) semen diluents, restrain, blocking of hog cholera virus and porcine reproductive and respiratory syndrome virus spread through artificial insemination.After the comparison test and production practice, suitable for boar semen diluents composition:each1000ml double distilled water, glucose15.50g, tri-sodium citrate11.65g, EDTA2.35g, sodium bicarbonate1.75g, polyvinyl alcohol (PVP, Type11)1.00g, three carboxyl methyl aminomethane (Tris)5.50g, citric acid4.10g, cysteamine acid0.07g, vitamin C1.0g, enrofloxacin0.1g, miconazole0.1g, Yinhuang and Ribavirin each of0.01g.The efficient semen diluents used in pig production and achieved fine results. Through the practical application of pig enterprises efficient semen technology experiment, implementation of semen in tropical regions of constant temperature preservation for6days, the average sperm activity was0.7, granted with breeding sow estrus pregnancy rate was86%, the average litter size of11.2postpartum, piglet mortality by13%, herd health rate increase32%in average, efficient effect and semen pig enterprise original common dilution liquid, improve overall efficiency of15%. Efficient semen diluents used to obtain good effect in practical application in pig artificial insemination.The first set of technical standards in Hainan enterprises pig artificial insemination. Efficient semen technology achievement maturity high, generate economic benefits4086600Yuan in the industry of the farm pig semen and comprehensive application of efficient, profit of2316200Yuan, effectively improve the comprehensive benefit of the enterprise culture.Popularization and application of technology of porcine efficient semen diluents with feeding, breeding, environmental control, prevention and information in one, in Hainan province and even the national level in the lead of technology. Hainan province existing300000basic sow, years of semen demand in1500000(80ml/bottle), porcine efficient semen dilution will rapidly popularized and applied in the province and achieved good social and economic benefits. Along with the scale pig farms and pig producers to further understanding of the science and technology, Hainan swine enterprises demand for fine varieties, integrated technology, the new technology and new equipment, improve. Therefore, popularization and application of efficient semen in Hainan province to carry out scientific pig played a demonstration and promotion role produced good social and economic benefits and disease control, ecological benefit. With the further development of the pig industry, social demand for high-end healthy pork products continue to improve, efficient semen dilution will be further applied to the pig industry, improve the level of production technology industry in china.
