Studies On The Immune Behaviour In Cell And Cytokin During The Havey Load Resistance Training

This study aimed to determine the effect of heavy load resistance training (HLRT)on the function of immunity and endocrine especially the change of molecular and cellular immunity in athletes, and to discovery some sensitive paramaters which indicated the physical condition of athletes during training. The study included two parts.partⅠ. Studies on the parameter changes immunity,endocrine and hemogram during the havey load resistance trainingFourteen lite male weight-lifters were enlisted in a 5-week weight-lifting training program which involved double 2-week training of heavey load of 5-8RM, and 30-50 day with an interval of an week adjusting ,Ten adult men were enlisted as control without resistance training. the blood sample were collected to measure CK, BUN, T, C, LH, FSH, PRL, RBC, Hb, Ret, lymphocyte subset(CD3~+, CD4~+, CD8~+, CD14~+, CD16~+, CD33~+, CD56~+, HLA-DR~+, Vα-24,Vβ-11 etc.), and the gene expression of IL-2, IL-4, IL-10 and IFN-γin peripheral leucocytes which were assessed by Real Time TR-PCR and quantitive measurement of mRNA.There is a significant increase in the level of CK and BUN, a slight decrease in T and a slight increase in C, a decrease tendency in T/C, and a mild fluctuation in LH and FSH during the both sessions of 2-week large volume training, while there existed a tendency to the basal level in the session of 1-week small volume training. The level of Hb decreased continuously until the end of the session of 1-week small volume training and returned to the basal level at the end of the test. The level of Ret increased significantly. The percentage of CD8~+, CD14~+, CD16~+, CD33~+, CD56~+, and Vα-24increased in the whole session, while the level of HLA-DR~+ increased only in the first session of 2-week. CD4~+/CD8~+ decreased significantly in the first session of 2-week and increased gradually then. There is a continuous decrease in the mRNA expression of IL-10, while that of IL-2, IL-4 increased first at first training session then decreased in the rest time. The mRNA expression of IFN-γdecreased significantly in the two sessions of large volume training, and increased significantly in the small volume session, the same as IFN-γmRNA/IL-4 mRNA.There were no significant changes in controls.Conclusion1. havey load resistance training led to a significant increase of the level of CK and BUN.2. There was a increase in the level of C, a decrease in T; a increase in the level of LH, a decrease in FSH during a large volume resistance training.3. havey load resistance training led to a transiently decrease in the level of RBC and Hb, but the level of Ret increased significantly, which means there was no inhibition of hematopoiesis during the whole session of training.4. havey load resistance training led to the level of CD8~+ increased significantly, CD4~+/CD8~+ decreased significantly in the first session of 2-week and increased gradually in the second 2-week session, and the mRNA expression of IL-2, IFN-γ, IFN-γ/IL-4 decreased as the training load increased, suggesting these are sensitive indexes that indicated the imbalance of immunity, the decrease of the anti-virus ability and the secretion function of T cells.5. IFN-γmRNA/IL-4 mRNA partly indicated Th1/Th2 since IFN-γcame mainly from Th1 cells, and IL-4 came from Th2 cells mainly. Large volume training led to a imbalance of Th1/Th2 and moved to Th2.TestⅡ. The function of cellular and molecular immunity changing in the pre-season training Fourteen male National Volleyballers were massed in 2 months'pre-season training.blood samples were colected ted before ,in the middle of training and in the end of training to measure CK, BUN, RBC, Hb, leucocytes, CD3~+, CD4~+, CD8~+, CD14~+, the gene expression of IL-2, IL-4, IL-10 and IFN-γin peripheral leucocytes which were assessed by Real Time TR-PCR and quantitive measurement of mRNA.The result indicated that the level of CK and BUN decreased, the level of RBC and Hb remained. There were no significant changes among leucocytes, CD3~+, CD4~+, CD8~+, CD4~+/CD8~+, while there was a decrease in the level of CD14~+. There was no significant change in the mRNA expression of IL-2 before and after the training, a increasing tendency in IFN-γmRNA, but there was a significant decrease in the mRNA expression IL-4, a significant increase in IL-10 mRNA, and a significant increase in IFN-γ/IL-4.Conclusion1. Training led to a decrease of CK and BUN, suggesting a recovery of athletes'exercise performance.2. Pre-season training led to a increasing tendency in the mRNA expression of IFN-γ, a significant decrease in IL-4, a significant increase in IFN-γmRNA /IL-4 mRNA, and a significant increase in IL-10. The result suggested that these indexes were sensitive to indicate the imbalance of immunity, the decrease of the anti-virus ability and the secretion function of T cells.3. The balance of Th1/Th2 moved to Th1.
