| The purpose of this study was to explore the mechanism of glycogen recovery after exercise and glucose and/or acanthopanacis senticosi supplement promoting skeletal muscle glycogen synthesis after exercise. We were also intent to provide a theoretic gist of promoting the recovery of athlete, elevating their training effect and augmenting their muscle glycogen synthesis.To achiever this purpose, a mechanism study experiment was arranged after animal model established by preliminary experiment. The mechanism study experiment adopted a week training mode with time and intension change so as to be close to the fact of athlete training. Subsequently, all rats completed glycogen depletion exercise (combined with high intensity /short time exercise and endurance/long time exercise). All rats should be killed after glycogen depletion exercise(before exercise,0h,4h and 12h after exercise, respectively).The value of serum glucose(SG), muscle glycogen, liver glycogen, insulin, insulin receptor, glucagons, GLIT4, GS, HK and AMPK were analyzed.Result discover:①Glucose supplement could extent the rat swimming time to exhaution . Glucose and acanthopanacis senticosi supplement had a cooperate function to promote the antifatigue ability.②The exhaustion of the glycogen in muscle and liver by exercise was recovered after exercise gradually. Supplement of glucose or acanthopanacis senticosi can promote the recover speed of muscle glycogen.③Glucose supplement was benefit to maintain the serum glucose level after exercise and promoted the liver glycogen recover. However, acanthopanacis senticosi supplement had little effect on the liver glycogen recover.④Glucose supplement could elevate the serum insulin level without effect on the insulin receptor. Acanthopanacis senticosi had little effect on the serum insulin level but could raise the content of insulin receptor.⑤Both Glucose supplement and acanthopanacis senticosi supplement could cut the level of serum glucagons and the preferably effect should emerge if glucose and acanthopanacis senticosi were supplied at the same time.⑥The GLUT4 transposition should increase after exercise. Glucose supplement is helpful for the transposition of GLUT4 from intracellular membrane to extraneous coat. On the other hand, acanthopanacis senticosi supplement should not only promote the transposition of GLUT4 from intracellular membrane to extraneous coat but also augment the total GLUT4 protein content.⑦After exercise, the activities of GS and HK were rise. There was little effect on those enzymes by glucose supplement. However, acanthopanacis senticosi supplement could strengthen the activity of those enzymes.⑧After exercise, the protein content of AMPK should increase. Glucose supplement can partly depress the protein contein. However, acanthopanacis senticosi supplement increase the protein content of AMPK.Our study indicated: glucose and/or acanthopanacis senticosi supplement could promote the muscle glycogen synthesis, but their mechanism were different.The glucose supplement mostly added the substance accommodate, elevated the level of serum glucose, stimulated the insulin excrete and increased the GLUT4 transposition. However, the acanthopanacis senticosi supplement promoted the sensitivity to insulin, increased the GLUT4 quantity as well as transposition, also enhanced the AMPK quantity to augment the glucose incept in muscle cell and heighten the GS and HK ability. |
