Objective:To study the effect of ultramicrostructure of brain tissue and the ERK signal transduction pathway and neural stem cells proliferation and differentiation in brain ischemia rat treated with moxibustion at Ren meridians.Methods:SD rats,Male,were divided into 4 groups randomly:Normal control group, Sham Operation group,MCAO group,Moxibustion Ren Meridians group(MR).8 rats in Normal group,24 rats in the other groups were averagely divided into 7 days group,14 days group and 28 days group at random.Cerebral ischemia-reperfusion injury model was induced by the method of nylon monofilament embolism.The ultramicrostructure changing of ischemic lateral cerebral corpus striatum and hippocampus in 7 days of Sham Operation group,MCAO group and Moxibustion Ren Meridians group were observed by electron microscope.ERK expression and phosphorylation on ischemic lateral cerebral ventricle and hippocampus dentate gyrus in MCAO rats were detected by Western Blot.BrdU positive cell and BrdU/Nestin double labeled cell expression on ischemic lateral cerebral ventricle, GFAP positive cell and GFAP/NSE double labeled cell expression on ischemic hippocampus dentate gyrus were detected by immunocyleochemistry method,laser confocal microscopy and fluorescence microscopy.Results:1,To compared with homeochronous MCAO rats(7 days after cerebral ischemia ),the ultramicrostructure of ischemic lateral cerebral tissue could be changed better by moxibustion at Ren meridians. 2,Compared with MCAO group,the ERK1/2 expression on ischemic lateral cerebral ventricle and hippocampus dentate gyrus in MR group was boosted after 14 days. Compared with MCAO group,the ERK1/2 phosphorylation in ischemic lateral cerebral ventricle and hippocampus dentate gyrus in MR group was significantly boosted after 7 days.It implies that moxibustion Ren Meridians can activate the ERK signal transduction pathway in ischemic lateral cerebral ventricle and hippocampus dentate gyrus,contributing to the differentiation and proliferation of the cells.3,The BrdU expression enhanced,while the number of BrdU/Nestin labeled cells accrescenced in MR group on ischemic lateral cerebral ventricle.It implies that the proliferation of neural stem cells on ischemic lateral cerebral ventricle were enhanced by moxibustion at Ren meridians.4,Moxibustion at Ren meridians could attenuate the GFAP expression on ischemic lateral hippocampus dentate gyrus,while increased the BrdU/Nestin double labeled cells after ischemia.It implies that moxibustion Ren Meridians can restrain the excess proliferation of hippocamp horizontal cells after ischemia while enhance the cell differentiation.Conclusions:The ultramicrostructure of ischemic lateral cerebral tissue could be changed better by moxibustion at Ren meridians,which can activate the ERK signal transduction pathway in MCAO rats and can promote the proliferation and differentiation of neural stem cell in MCAO rats.
