Research Objects This dissertation is intended to do a pioneering research by meansof literature review of theoretical knowledge and modern researchdevelopment on treating ischemic stroke in traditional Chinese medicineand modern medicine.It is believed that qi deficiency and blood stasis is a main pathogenesisof ischemic stroke based on it.By experiments it is systematicallyobserved how Yiqi Huoxue therapy affects ischemic stroke at differentlevels, different points and various stages,its mechanism isillustrated by molecule biology in View of signal transduction pathwayand modulation of nerve cell apoptosis,at the same time the experimentsreveal the pathogenic essence of qi deficiency and blood stasis ofischemic stroke.We endeavor to provide scientific theory andexperimental evidence for effecfively treating ischemic stroke onclinic basis and developing modern Chinese medicine.Main Research Idess Ischemic stroke is a dynamic process of various factors,links,ways,points and taches because of its complexity and variety .Since Chinesemedical treatment aims to improve and protect ischemia/reperfusion(I/R)injury at different levels different ways and different points,thisresearch observe systematically that Yiqi Huoxue therapy on apoptosisafter I/R injury at different levels and stages based on a mainpathogenesis-qi deficiency and blood stasis.In Fas/FasL signaltransduction pathway and modulation of nerve cell apoptosisinterrelated Anti-apoptosis gene expression and pro-apoptosis geneexpression are observed in ischemic penumbra(IP) area.Therefore moleculebiological mechanism is explored that Yiqi Huoxue therapy can restrainneuron apoptosis in I/P area of animal model of qi deficiency and bloodstasis syndrome with local cerebral.Main Method Grangness investigation method :mortality rate,weight,neurologicalsymptom ,qi difficiency symptom,blood stasis symptom .Microcosmicinvestigation method:cAMP and cGMP contents in the plasma and cerebraltissue, cerebral tissue water content, HE,Thionine thionin staining,Marchalls staining, Mallory staining, immunohistochemistry staining and insitu hybridization.Dynamic investigation method:SD rats of aboat 300-320g are made into the qi deficiency and blood .stasis model by meansof combined factors of hunger,tiredness and high fat diet and 1ater aremade into local cerebral I/R model by thread-occluding the middlecerebral artery(MCA).These test rats are randomly divided into pseudosurgery group ,model group, yiqi group,huoxue group and yiqi Huoxuegroup after reperfusion 1,3 and 7 days following focal cerebral ischemiafor 2 hours.The combination of micro and macro research,qualitafive andquantitative research,dynamic and static observation,therapy,syndromeand formula and Yiqi,Huoxue and YiqiHuoxue therapy comparative researchare used.Mathematic investigation method :Metamorph Imagine AnalysisSystem and SPSS 11.0 for Windows are applied.Results1 The effect of Yiqi Huoxue therapy on mortality rate of model rats The results show that mortality rate of yiqi group and yiqi huoxuegroup is apparently higher than model group and huoxue group at thesame stage.2 The effect of Yiqi Huoxue therapy on weight of model rats The groups after surgery varies in the decrease of weight of modelrats except pseudo surgery group.After treatment all groups increase inweight,there is significant difference in weight increase between themodel group and Yiqi Huoxue group at the same stage(7d).3 The effect of Yiqi Huoxue therapy on neurological symptom of model rats The experiment show that causes rats severe neurological deficiencyafer making local cerebral I/R model by threed-occluding the middlecerebral artery(MCA).
