| Forsythia suspensa (Thunb) Vahl. (Oleaceae) is an important original plant of the Chinese traditional drug "lianqiao" which had been used for antiinflammatory, antibacterial, diuretic, drainage and antidotal purposes in traditional Chinese medicine. In order to search for the biologically active substances, we have reinvestigated the chemical constituents of F. suspensa. 36 compounds, including caffeoyl glycosides, cyclohexylethane derivatives, flavonoides, iridoid glycoside, lignans, and triterpenes etc., have been isolated from the 70% EtOH extract of this plant, among which 6 were new compounds, named as (+)-suspensaside A (1), suspensaside B (2), suspenolic acid (5), forsythenside A (6), forsythenside B (7), and forsythenin (14). Their structures were elucidated by means of various modern spectroscopic techniques, X-ray and chemical methods. The known compounds were forsythiaside (3), suspensaside (4), rengyol (8), rengyoside A (9), rengyoside B (10), rengyoside C (11), salidroside (12), rengyolone (13), phillygenin (15), pinoresinol (16), phillyrin (17), (+)-pinoresinol-P-D-glucoside (18), isobaueryl acetate (22), P-amyrin acetate (23), 20(s)-dammar-24-ene-3p,20-diol-3-acetate (24), betulinic acid (25), ursolic acid (27), oleanolic acid (28), P-sitosterol (29), rutin (31), (6'-o-palmitoyl)-sitosterol-3-o-P-D-glucoside (19), ocotillone (20), ocotillol monoacetate (21), adoxosidic acid (26), daucosterol (30), caffeic acid (32), vanillic acid (33), p-hydroxyphenyl acetic acid (34), succinic acid (35), and palmitic acid (36). The latter 10 of compounds were obtained from this species for the first time.The pharmacological screens in vitro disclosed that the principal antibacterial constituents from F. suspensa were caffeoyl glycosides. The results of antitumor test in vitro against human tumor cell line showed that betulinic acid exhibited a weak activities to the human Leukaenia (HL-60) and L1210 cells. The inhibiting rates of... |
