Chinese medicine formula is the main form of preventing and treating diseases in the clinic of Chinese medicine. The study of Chinese medicine compounds have become the highlight of the study of Chinese medicine. The biological effect model of Chinese medicine compound acting on the human body is one of the key problems in the study of Chinese medicine compound mechanism. To explore the unique effect model of Chinese medicine compound taking advantage of the superiority of compatibility and the character of multicomponents, multitargets and multi-pathway regulation is of very importance for the study of Chinese medicine theory and the application of the Chinese medicine in the clinic.This topic takes the hepatocyte injury induced by CCl4 as the entrance, and the hepatocyte as the carrier, the Chinese medicine compound is simplified to several monomers, the hepatoprotective effects of these monomers and the compatibilities of them which are below the threshold concentrations are investigated systematically in cell and mice.This dissertation is divided into two parts. In the literature part, the mechanism of the Chinese medicine compound is summarized, the pharmacologic actions of the four monomers used in the experiment which are baicalin, quercetin, eugenol, and linalool are reviewed. In the experiment part, the stable hepatocyte injury model is established, the effect threshold concentrations of the four monomers are found, the effects of compatibilities of the four monomers which are combined according to the orthogonality experiment design are investigated, the influences of the compatibilities of the four monomers on the results of the drug effects are analyzed to presume the possible biological effect model. And then dose and effect relationship of the best formula is reviewed, the curative effect of the optimized monomers formula under the condition of low concentration on the mice liver injury model is tested.Study one The establishment of the hepatocyte injury model made on BRL-3 A rat hepatocyte induced by CCl4Methods: rat hepatocytes are divided into control group, 5mmol/L CCl4 group, 10mmol/L CCl4 group, 15mmol/L CCl4 group, 20mmol/L CCl4 group, the culture medium without serum is added into the control group cells, the CCl4 with corresponding concentration is added into cells. Then, the clear supernatant liquid in the hole of the cell are withdrawn after they have been cultured in 3h, 6h, 12h, 24h, 48h, the ALT levels are measured. The cell multiplication rates are measured by using the rest cells through theMTT method. Results: compared with the control group, the ALT levels of the 5mrnol/L CCl4 group, 10mmol/L CCl4 group, 15mmol/L CCl4 group, 20mmol/L CCl4 group acted by CCU after 3h, 6h, 12h were increased significantly, P<0.05. the ALT levels of 10mmol/L CCU group, 15mmol/L CCU group, 20mmol/L CCU group were increased significantly also, P<0.05. Compared with the control group, the ALT level of the 5mmol/L CCU group acted by CCU after 24h had not increased significantly, P>0.05. The ALT level of 5mmol/L CCU group, 10mmol/L CCU group, 15mmol/L CCU group, 20mmol/L CCU group acted by CCU after 48h had not increased significantly, P>0.05. Compared with the control group, the hepatocyte proliferation rates of 5mmol/L CCU group, 10mmol/L CCU group, 15mmol/L CCU group, 20mmol/L CCU group acted by CCU after 3h, 6h, 12h, 24h had no significant change. The hepatocyte proliferation rates of 5mmol/L CCU group, lOmmol/L CCU group acted by CCU after 48h increased significantly, P<0.05. 15mmol/L CCU group, 20mmol/L CCU group acted by CCU after 48h had no significant change, P<0.05.
