Crohn's disease (CD) and ulcerative colitis(UC), the two main forms of inflammatorybowel disease (IBD), are characteristically chronic diseases resulting from the combinedcontribution of environmental, genetic, bacterial, and immune factors. The role of immunefactors is becoming more and more important. It's difficult to treat and there are no knowncures at the present. Current treatment strategies endeavor to control the underlyinginflammation, and conventional anti- inflammatory agents are used widely in this regard. According its clinical symptoms, IBD belongs to "diarrhea" and "dysentery" intraditional Chinese medicine (TCM). SISHENFANG (SSF),XIANGLIANFANG (XLF),CHAGNTAIFANG (CTF) has the function of warming the kidney and relieving diarrhea,clearing away heat evil and eliminating dampness evil, warming the kidney and clearingaway and eliminating evil respectively. In order to know whether the effect and mechanismare different between the three prescriptions, experimental colitis models (including TNBSmouse and DSS mouse) were used. The establishing of experimental colitis models To establish the colitis model, female Balb/c mice received enemas of 1,1.5,2mgTNBS in 50% ethanol. The weight, death rate, diarrhea, DAI score, colon macrophage andmicrophage histological score, MPO were observed. The result suggest that 1.5mg TNBSin 50% ethanol for each mouse is enough to establish experimental colitis model. Furtherresearch indicates that 10 days after the enema, evident surgery is still exist in mice'scolon. DSS was dissolved in water to provide a working solution of 3% (w/v) for oralconsumption by C57BL/6 mice for 5 days, then the DSS solution was replaced by water. 3days later mice were killed. Fluctuations in body weight, stool consistency and blood lossfrom the anus were monitored throughout the experimental period. According the histological characters, mice with TNBS colitis are remarkably similarto human's CD; on the other hand, mice with DSS colitis are similar to UC. The effect of CTF,SSF,XLF on IBD animal models Experimental colitis was induced by TNBS and DSS respectively, and its severity wasevaluated by gross colon injury, histological damage, body weight, survival rate, and theproduction of MPO. We studied the effect of CTF,SSF,XLF on all these parameters. Theresults indicated that CTF,SSF,XLF significantly suppressed the various manifestations ofexperimental colitis as demonstrated by substantial reduction in the macroscopic colonicdamage, preservation of the microscopic colonic structure, reduced weight loss and MPO,and improved long-term survival. It suggest that they all can prevent induction of and treatTNBS colitis, and the high,middle dose of the CTF are more effective compared withSASP, a classical medicine for IBD. SSF's effect is equal to CTF, while XLF has less effecton improve survival rate or reduce weight loss. Study on mechanisms of CTF,SSF,XLF on IBD in TNBS colitis The mice colitis model was induced by TNBS. The costimulatory molecular surfacedisplay, the rate of sub-T cell, the secretion of cytokines were observed to detect themechanisms of affection of CTF,SSF,XLF on TNBS colitis.1. the effect on activating of T cell Secretion of IL-12/23 was determined by ELISA, the costimulatory molecule surfacedisplays were determined by FACS analysis and the proliferation of T cell were determinedby MTT. Through observating the effect of the three prescriptions on these parameters, theactivating of T cells was evaluated. The results show that there are notably activation of Tcells in TNBS colitis, which may through improving the CD40-CD40L and the B7(CD80/CD86) - CD28/CTLA4 pathway, and the secretion of IL-12/23 were higher thancontrol group. The three prescriptions all can inhibit the surface display of thecostimulatory molecule, inhibit the activation of T cells, and suppress the secretion ofIL-12/23. At the same time, they can also improve the proliferation of T cell, and theeffects of CTF and SSF are more remarkable. 2. the effect on the CD4+ T cell's...
